人口腔疣狀癌裸鼠移植瘤模型的建立及Survivin和Bcl-2的表達分析

【摘要】： 目的:對口腔疣狀癌的裸鼠建模進行初步的實驗研究,為深入研究口腔疣狀癌的發(fā)生、發(fā)展及其演變機理提供有益的探索模型。 材料與方法:將口腔疣狀癌癌組織采用組織塊移植法和細胞懸液注射法移植于Balb/c裸鼠皮下,建立人口腔疣狀癌裸鼠移植瘤模型(模型一);將復(fù)發(fā)性疣狀癌伴鱗癌頸部轉(zhuǎn)移灶采用組織塊移植法移植于裸鼠右后背部皮下,建立人復(fù)發(fā)性疣狀癌術(shù)后伴鱗癌頸淋巴轉(zhuǎn)移裸鼠移植瘤模型(模型二)。觀察移植瘤生長特性,計算移植成功率,繪制生長曲線,并對移植瘤進行病理學和組織學檢查。 結(jié)果:模型一建模失敗,模型二原代移植于8只裸鼠有7只獲得成功,成瘤率87.5%(7/8),腫瘤生長曲線符合Gompertz函數(shù)曲線的一般規(guī)律。移植瘤最后都出現(xiàn)壞死,個別還出現(xiàn)消退現(xiàn)象。皮下腫瘤瘤體呈圓球形或橢圓形,表面光滑,包膜完整,呈結(jié)節(jié)狀突起,與周圍組織界限清楚,活動度良好,有的腫瘤呈向心性壞死,有的腫瘤表面可見清晰的毛細血管,表面肉紅色,剖面呈魚肉狀,質(zhì)韌,有的呈白色豆渣狀。光鏡下移植瘤細胞成多角形,異型性明顯,核漿比例增大。核橢圓形、圓形、大小不等,核分裂相多見,并見病理核分裂。癌細胞排列呈巢狀、片狀或條索狀,其間可見角化珠。肝、肺、腎未見轉(zhuǎn)移,前肢腋下及腹股溝淋巴結(jié)反應(yīng)性增生。移植瘤中Survivin,Bcl-2蛋白免疫組織化學染色和人腫瘤比較差異無顯著性(P＞0.05)。 結(jié)論: 1.建立人復(fù)發(fā)性疣狀癌術(shù)后伴鱗癌頸淋巴結(jié)轉(zhuǎn)移裸鼠原代移植瘤模型獲得成功,為下一步建立穩(wěn)定的移植瘤模型奠定了堅實的基礎(chǔ)。 2.人復(fù)發(fā)性疣狀癌伴鱗癌頸淋巴轉(zhuǎn)移原代移植瘤保留了人鱗癌的生物學特點,并與原發(fā)腫瘤的組織學特征相似。 3.Survivin和Bcl-2在移植瘤及人高分化鱗癌中的表達差異無顯著性。
[Abstract]:Objective: To study the model of oral cavity carcinoma in nude mice, and to provide a useful exploration model for the study of the occurrence, development and evolution mechanism of oral cavity carcinoma. Materials and Methods: The tissue-block transplantation method and cell suspension injection method were applied to the subcutaneous of Balb/ c nude mice. 1). The cervical lymph node metastasis of recurrent cervical carcinoma with squamous cell carcinoma was transplanted to the right back of nude mice by a tissue-block transplantation method, and a model of the tumor-associated cervical lymph node metastasis in nude mice (model) was established. 2) observing the growth characteristics of the transplanted tumor, calculating the success rate of the transplantation, drawing the growth curve, and carrying out pathology and histology on the transplanted tumor. Results: The model one model failed, the model 2 was transplanted to 8 nude mice with 7 success, the tumor rate was 87.5% (7/8), and the tumor growth curve was in line with the Gompertz function curve.
