小鼠死后滯留對(duì)卵巢卵母細(xì)胞存活與成熟的影響

【摘要】:目的本實(shí)驗(yàn)為了探討不同的卵母細(xì)胞非離體存放時(shí)間和存放的環(huán)境溫度變化對(duì)小鼠卵巢內(nèi)卵母細(xì)胞存活與體外成熟的影響 方法8周齡的ICR品系雌鼠隨機(jī)分組,每組5只。利用二氧化碳安樂(lè)死方法處死后將尸體存放在室溫(25℃),低溫(4℃)和高溫(37℃)環(huán)境里一定時(shí)間(0,2,4,6,8和10h)后摘取卵巢組織剁碎后收集生發(fā)泡(GV)期卵母細(xì)胞。隨后對(duì)收集到的卵母細(xì)胞進(jìn)行形態(tài)學(xué)檢測(cè),并統(tǒng)計(jì)每組GV期卵母細(xì)胞的數(shù)目,同時(shí)對(duì)其體外培養(yǎng)過(guò)程中的成熟率以及后續(xù)卵母細(xì)胞染色體和紡錘體還有細(xì)胞質(zhì)中線粒體的分布和谷胱甘肽(GSH)含量也進(jìn)行檢測(cè)。 結(jié)果1室溫(25℃)下小鼠尸體可以存放至4小時(shí)并收集到GV期卵母細(xì)胞,低溫(4℃)環(huán)境中可以存放至10小時(shí),而高溫(37℃)組中只能存放1小時(shí)。2我們統(tǒng)計(jì)了不同條件下GV期卵母細(xì)胞的數(shù)目,結(jié)果顯示在同一溫度下,隨著存放時(shí)間延長(zhǎng),收集到的GV期卵母細(xì)胞總數(shù)目隨之減少。3在存放相同時(shí)間的情況下,收集到的GV期卵母細(xì)胞體外培養(yǎng)成熟過(guò)程中其生發(fā)泡破裂(GVBD)比率和第一極體(PB)排出比率也隨著存放溫度的升高而降低。4結(jié)果發(fā)現(xiàn)各實(shí)驗(yàn)組中細(xì)胞質(zhì)內(nèi)不正常線粒體的分布比率遠(yuǎn)遠(yuǎn)高于對(duì)照組,但是卵母細(xì)胞細(xì)胞質(zhì)中GSH含量并沒有明顯差異。5實(shí)驗(yàn)組中卵母細(xì)胞內(nèi)染色體排列和紡錘體組裝過(guò)程都與對(duì)照組一致。 結(jié)論1小鼠死后滯留影響了GV期卵母細(xì)胞的存活數(shù)目。2小鼠死后滯留的低溫環(huán)境可以延長(zhǎng)尸體的存放時(shí)間,,但是高溫環(huán)境則減少存放時(shí)間。3小鼠死后滯留影響了卵母細(xì)胞體外成熟程度,并延遲卵母細(xì)胞的成熟進(jìn)程。4小鼠死后滯留造成卵母細(xì)胞內(nèi)不正常線粒體分布比率上升。5小鼠卵母細(xì)胞染色體,紡錘體和谷胱甘肽含量在一定時(shí)間內(nèi)不受死后滯留時(shí)間影響。
