【摘要】： 目的:本文研究降鈣素基因相關(guān)肽(CGRP)對HaCaT細胞增殖、凋亡及分泌NGF的影響。 方法:以不同濃度(10~(-10)mol/L～10~(-7)mol/L)CGRP作用于HaCaT細胞,用MTT法檢測對細胞增殖及生長曲線的影響;流式細胞術(shù)分析對細胞凋亡的影響;酶聯(lián)免疫吸附測定(ELISA)法檢測細胞上清液中NGF濃度的變化。應(yīng)用SPSS 13.0統(tǒng)計軟件包進行統(tǒng)計學(xué)處理,差別有顯著性標(biāo)準(zhǔn)為P＜0.05。 結(jié)果:CGRP促HaCaT細胞增殖作用的強度與CGRP的濃度和作用時間有關(guān):CGRP濃度為10~(-10)mol/L至10~(-8)mol/L時,其促增殖作用呈濃度依賴性(F=7.824 P＜0.01);當(dāng)濃度上升至10~(-7)mol/L時促增殖作用較10~(-8)mol/L時減弱(F=15.231 P＜0.01);CGRP作用48小時促增殖作用最強;流式細胞術(shù)分析發(fā)現(xiàn)隨CGRP濃度升高,其抑制HaCaT細胞凋亡的作用增強;ELISA檢測發(fā)現(xiàn)HaCaT細胞上清液中NGF濃度隨CGRP濃度升高和作用后時間延長而增加(F=32.207、76.520 P＜0.01)。 結(jié)論:CGRP可促進HaCaT細胞增殖、抑制其凋亡,并可促進HaCaT細胞分泌NGF。HaCaT細胞分泌NGF增加可能是CGRP促進增殖、抑制凋亡的原因之一。
[Abstract]:Aim: to study the effects of calcitonin gene-related peptide (CGRP) on the proliferation, apoptosis and secretion of NGF in HaCaT cells. Methods: HaCaT cells were treated with different concentrations (10 ~ (-10) mol/L~10~ (-7) mol/L) CGRP, and the effects on cell proliferation and growth curve were detected by MTT assay, and apoptosis was analyzed by flow cytometry. Enzyme linked immunosorbent assay (ELISA) was used to detect the concentration of NGF in supernatant. SPSS 13.0 statistical software package was used for statistical analysis, and the difference was significant (P < 0.05). Results: the effect of CGRP on the proliferation of HaCaT cells was related to the concentration and time of CGRP. When the concentration of CGRP was from 10 ~ (-10) mol/L to 10 ~ (-8) mol/L, the effect of CGRP on proliferation of HaCaT cells was concentration-dependent (FF7. 824 P < 0. 01). When the concentration increased to 10 ~ (-7) mol/L, the effect of promoting proliferation was weaker than that of 10 ~ (-8) mol/L (F _ (15.231) P < 0. 01); CGRP). Flow cytometry showed that the inhibitory effect of CGRP on apoptosis of HaCaT cells was enhanced with the increase of CGRP concentration, and that the concentration of NGF in the supernatant of HaCaT cells increased with the increase of CGRP concentration and the prolongation of the time after treatment by ELISA assay (FN 32.207? 76.520 P < 0. 01). Conclusion: CGRP can promote the proliferation and inhibit the apoptosis of HaCaT cells and increase the secretion of NGF by NGF.HaCaT cells of HaCaT cells which may be one of the reasons of CGRP promoting proliferation and inhibiting apoptosis.
