1型糖尿病患者外周血漿細(xì)胞樣樹突狀細(xì)胞數(shù)量及功能的研究

【摘要】: 目的:分析1型糖尿病患者外周血漿細(xì)胞樣樹突狀細(xì)胞(PDC)數(shù)量及功能變化。 方法:采用流式細(xì)胞分析技術(shù)檢測22例1型糖尿病患者(T1DM)及20例健康人體內(nèi)PDC細(xì)胞的比例及數(shù)量;用免疫刺激序列(CpG ODN-2216)刺激外周血單個核細(xì)胞(PBMCs)并與PBMCs共培養(yǎng),通過檢測PBMCs產(chǎn)生的IFN-α水平評估PDC的功能。 結(jié)果:與健康對照組比較,1型糖尿病患者組外周血PDC的百分比和數(shù)量均顯著下降(p<0.01);PDC經(jīng)CpG ODN-2216刺激后產(chǎn)生IFN-α的能力較健康對照組也有顯著下降(p<0.05);相關(guān)性分析表明,經(jīng)CpG ODN-2216刺激后IFN-α的產(chǎn)量與PDC數(shù)量呈正相關(guān)關(guān)系(r=0.439,p<0.05)。 結(jié)論:1型糖尿病患者體內(nèi)PDC數(shù)量和功能異常,說明PDC可能積極參與了患者機(jī)體的免疫紊亂過程;PDC產(chǎn)生的IFN-α減少,可能使患者對感染的控制能力降低而易感。
[Abstract]:Objective: to analyze the changes of (PDC) number and function of plasma cell like dendritic cells (PBMC) in type 1 diabetes mellitus (DM). Methods: the proportion and quantity of PDC cells in 22 patients with type 1 diabetes mellitus (T1DM) and 20 healthy people were detected by flow cytometry. Peripheral blood mononuclear cells (PBMCs) were stimulated by immunostimulatory sequence (CpG ODN-2216) and co-cultured with PBMCs. The function of PDC was evaluated by detecting the level of IFN- 偽 produced by PBMCs. Results: compared with the healthy control group, the percentage and quantity of PDC in peripheral blood of type 1 diabetic patients decreased significantly (p < 0. 01). The ability of PDC stimulated by CpG ODN-2216 to produce IFN- 偽 was significantly lower than that of control group (p < 0. 05). Correlation analysis showed that the yield of IFN- 偽 was positively correlated with the quantity of PDC after CpG ODN-2216 stimulation (r = 0. 439, p < 0. 05). Conclusion: the abnormal number and function of PDC in patients with type 1 diabetes mellitus indicate that PDC may participate in the immune disorder process of the patients, and the decrease of IFN- 偽 produced by PDC may make the patients' ability to control infection lower and susceptible to infection.
