改良式液氮冷凍法制備犬股骨頭壞死動物模型的研究

【摘要】: 研究背景:股骨頭缺血壞死的治療是骨科臨床難題之一,目前尚未發(fā)現(xiàn)一種可終止或者逆轉(zhuǎn)已發(fā)生股骨頭壞死病理過程,從而阻止股骨頭進一步塌陷的有效方法。雖然我們已知在股骨頭壞死區(qū)的修復(fù)與重建過程中,壞死骨的吸收導(dǎo)致股骨頭強度的下降是塌陷的主要原因,減少或終止壞死骨的吸收可延遲或避免壞死股骨頭塌陷的發(fā)生。但我們對其發(fā)病機理和何種方法能有效治療股骨頭缺血性壞死尚不十分清楚,原因之一可歸咎于缺乏一個能真正反映股骨頭壞死的病理過程的動物模型。 目的:通過改良式液氮冷凍法造成犬股骨頭缺血壞死,探討股骨頭缺血壞死的動物模型制備,建立能模擬人股骨頭壞死(osteonecrosis of femoral head ,ONFH)病理生理過程的實驗動物模型。 方法:16只犬隨機分成4組,采用改良式液氮冷凍法即刻冷凍單側(cè)股骨頭,普通飼料飼養(yǎng)2、4、8、16周分別處死4組動物,分別觀察X線攝片、大體形態(tài)、光鏡下形態(tài)變化。 結(jié)果:改良式液氮冷凍法冷凍犬股骨頭X線、大體及組織學(xué)形態(tài)不同階段均出現(xiàn)明顯變化。 結(jié)論:改良式液氮冷凍法為建立股骨頭壞死動物模型提供了一種簡便、易行的方法,值得進一步深入研究。
[Abstract]:Background: the treatment of avascular necrosis of femoral head is one of the clinical problems in orthopedic department. At present, there is no effective method to stop or reverse the pathological process of femoral head necrosis and prevent the further collapse of femoral head. Although it is known that the reduction of the strength of the femoral head due to the absorption of the necrotic bone is the main cause of the collapse during the repair and reconstruction of the necrotic area of the femoral head, the reduction or termination of the absorption of the necrotic bone can delay or avoid the collapse of the necrotic femoral head. However, the pathogenesis and the effective treatment of avascular necrosis of femoral head are not well understood. One of the reasons can be attributed to the lack of an animal model which can truly reflect the pathological process of avascular necrosis of femoral head. Objective: to establish an experimental animal model of avascular necrosis of femoral head (ANFH) in dogs by using modified liquid nitrogen freezing method to establish an animal model of avascular necrosis of femoral head (ANFH), which can simulate the pathophysiological process of human avascular necrosis of femoral head (osteonecrosis of femoral head, ONFH). Methods: sixteen dogs were randomly divided into 4 groups, and the animals were killed for 16 weeks by using modified liquid nitrogen freezing method to freeze the unilateral femoral head immediately. The animals were killed for 16 weeks. The X-ray films, the gross morphology and the morphological changes under light microscope were observed. Results: there were obvious changes in X-ray, gross and histological morphology of femoral head frozen by modified liquid nitrogen freezing method. Conclusion: the modified liquid nitrogen freezing method provides a simple and easy method for the establishment of animal model of femoral head necrosis.
