去卵巢及雌激素替代治療后家兔邊緣系統(tǒng)NOS的活性及nNOS陽性神經(jīng)元的形態(tài)結(jié)構(gòu)與分布變化

【摘要】: 為了探討雌激素、孕激素對去卵巢家兔邊緣系統(tǒng)NOS的活性及對nNOS陽性神經(jīng)元形態(tài)結(jié)構(gòu)分布的影響,本試驗將家兔切除卵巢,作為婦女更年期后卵巢功能衰竭、雌激素分泌嚴(yán)重不足時的良好模型,然后通過人為注射雌激素及雌激素和孕激素,觀察去卵巢和注射雌激素及雌激素和孕激素后家兔血清中NO的濃度、邊緣系統(tǒng)NOS活性及nNOS陽性神經(jīng)元的形態(tài)結(jié)構(gòu)分布變化來研究雌激素保護中樞神經(jīng)系統(tǒng)、預(yù)防老年癡呆和記憶力減退等神經(jīng)系疾病的機理,為神經(jīng)生理學(xué)和病理學(xué)研究提供形態(tài)學(xué)依據(jù)。 利用SABC免疫組織化學(xué)法,在光學(xué)顯微鏡下對各組家兔海馬CA1、CA2、CA3區(qū)及齒狀回、下丘腦室旁核、下丘腦外側(cè)區(qū)、視上核以及額葉神經(jīng)型一氧化氮合酶(nNOS)陽性神經(jīng)元的形態(tài)結(jié)構(gòu)及分布進(jìn)行觀測。結(jié)果表明:假手術(shù)對照組家兔海馬、下丘腦、額葉內(nèi)有大量的nNOS陽性神經(jīng)元分布;去卵巢后nNOS陽性神經(jīng)元形態(tài)分布變化有區(qū)域差異,額葉和海馬各區(qū)(除CA2外)nNOS陽性神經(jīng)元表達(dá)明顯減弱,下丘腦各神經(jīng)核nNOS陽性神經(jīng)元的表達(dá)明顯增強,即數(shù)量增多,染色加深;雌激素替代治療組與假手術(shù)組之間無顯著差異,額葉和海馬各區(qū)(除CA2外)nNOS陽性神經(jīng)元表達(dá)明顯增強,下丘腦各神經(jīng)核nNOS陽性神經(jīng)元的表達(dá)明顯減弱,即數(shù)量減少,染色變淺。 利用硝酸還原酶法和生物化學(xué)檢測技術(shù),采用NO、NOS試劑盒分別檢測去卵巢和注射雌激素及雌激素和孕激素后家兔血清中NO的濃度的變化及大腦邊緣系統(tǒng)NOS的活性變化。結(jié)果表明:去卵巢三個月后家兔血清NO水平和大腦邊緣系統(tǒng)NOS活性降低;單純補充雌激素和聯(lián)合使用雌激素、孕激素時,血清NO水平和大腦邊緣系統(tǒng)NOS活性均升高;單純補充雌激素血清NO水平和大腦邊緣系統(tǒng)NOS的活性高于聯(lián)合使用雌激素、孕激素時血清NO水平和大腦邊緣系統(tǒng)NOS活性,但差異不顯著。
[Abstract]:In order to investigate the effects of estrogen and progesterone on the activity of NOS in the limbic system of ovariectomized rabbits and on the morphological and structural distribution of nNOS positive neurons, the ovariectomized rabbits were treated as ovarian failure after menopause. A good model when estrogen secretion was seriously deficient, then the serum NO concentration of rabbits after ovariectomized and injected with estrogen and progesterone was observed by artificial injection of estrogen, estrogen and progesterone. In order to study the mechanism of estrogen protecting the central nervous system and preventing senile dementia and memory loss, the activity of NOS in the limbic system and the morphological and structural changes of nNOS positive neurons were studied. To provide morphological basis for neurophysiology and pathology. Using SABC immunohistochemical method, the CA1,CA2,CA3 area, dentate gyrus, paraventricular nucleus of hypothalamus and lateral area of hypothalamus in each group were studied under optical microscope. The morphology and distribution of neuronal nitric oxide synthase (nNOS) positive neurons in supraoptic nucleus and frontal lobe were observed. The results showed that there were a large number of nNOS positive neurons in hippocampus, hypothalamus and frontal lobe of rabbits in sham operation control group, and there were regional differences in morphological distribution of nNOS positive neurons after ovariectomy. The expression of nNOS positive neurons in frontal lobe and hippocampal regions (except CA2) was significantly decreased, and the expression of nNOS positive neurons in hypothalamic nuclei was significantly increased, that is, the number of nNOS positive neurons increased and the staining deepened, there was no significant difference between estrogen replacement therapy group and sham-operation group. The expression of nNOS positive neurons in frontal lobe and hippocampal regions (except CA2) was significantly increased, and the expression of nNOS positive neurons in hypothalamic nuclei was significantly decreased, that is, the number of nNOS positive neurons decreased and the staining became shallower. NO,NOS kit was used to detect the changes of serum NO concentration and the activity of NOS in brain limbic system of rabbits after ovariectomized and injected estrogen and progesterone with nitrate reductase and biochemical techniques. The results showed that after ovariectomized for 3 months, the level of serum NO and the activity of NOS in the cerebrum limbic system were decreased, and the level of serum NO and the activity of NOS in the limbic system of the brain were increased after estrogen supplementation and combined use of progesterone. The level of serum NO and the activity of NOS in the limbic system of brain were higher than those in the combination of estrogen and progesterone, but there was no significant difference in the level of serum NO and the activity of NOS in the limbic system of brain with progesterone.
