凍干人用狂犬病脂質(zhì)體疫苗的研究

【摘要】: 人用狂犬病疫苗暴露前免疫程序一般為0、7、21或28天各接種1劑量的疫苗(2.5IU/劑),暴露后免疫為0、3、7、14、28天各接種1劑量的疫苗(2.5IU/劑)。該免疫程序的缺點(diǎn)是接種針次多,全程免疫時(shí)間長。 本文以脂質(zhì)體作為狂犬病疫苗的佐劑,制備了高效價(jià)的凍干人用狂犬病脂質(zhì)體疫苗(LipoRabV),目的是減少全程免疫針次,縮短全程免疫時(shí)間。本文以小鼠及比格犬為實(shí)驗(yàn)動(dòng)物對(duì)LipoRabV進(jìn)行了細(xì)胞免疫研究,體液免疫研究及疫苗保護(hù)效果研究。證明了脂質(zhì)體對(duì)狂犬病疫苗的佐劑作用,提高了疫苗的保護(hù)效力。在細(xì)胞免疫研究中,凍干人用狂犬病脂質(zhì)體疫苗促進(jìn)了特異性T淋巴細(xì)胞的增殖,促進(jìn)了細(xì)胞因子IL-2、IFN-γ的產(chǎn)生。體液免疫研究表明,LipoRabV 3針免疫產(chǎn)生的中和抗體不低于凍干人用狂犬病疫苗(RabV)5針免疫的中和抗體水平。在暴露后免疫程序研究中,LipoRabV 3針免疫保護(hù)率高于對(duì)照RabV5針免疫的保護(hù)率。 本文初步確定了凍干人用狂犬病脂質(zhì)體疫苗的免疫程序:暴露前免疫程序?yàn)?、14天各接種1劑量的凍干人用狂犬病脂質(zhì)體疫苗(6.0IU/劑);暴露后免疫程序?yàn)?、3、14天各接種1劑量的凍干人用狂犬病脂質(zhì)體疫苗(6.0IU/劑)。同現(xiàn)有疫苗相比,暴露前免疫程序由3針減至2針,全程免疫由28天減至14天。暴露后免疫程序由5針減至3針,全程免疫由28天減至14天。 本項(xiàng)目于2006年被立為國家“863”項(xiàng)目,項(xiàng)目編號(hào)為2006AA02A221。
[Abstract]:The routine of human rabies vaccine immunization before exposure was 1 dose of 2.5IU/ on 21 or 28 days after exposure, and 1 dose of 2.5IU/ was inoculated with 1 dose of 2.5IU/ on each day after exposure. The drawback of the immune program is that the vaccination times are many and the whole immunization time is long. Using liposome as adjuvant of rabies vaccine, a freeze-dried human rabies liposome vaccine (LipoRabV),) with high titer was prepared in this paper. In this paper, the cellular immunity, humoral immunity and vaccine protection of LipoRabV were studied in mice and Beagle dogs. The effect of liposome on the adjuvant of rabies vaccine was proved, and the protective effect of the vaccine was improved. In the study of cellular immunity, freeze-dried human rabies liposome vaccine promoted the proliferation of specific T lymphocytes and the production of cytokine IL-2,IFN- 緯. Humoral immunity studies showed that the neutralizing antibody produced by LipoRabV 3 doses was not lower than that of freeze-dried human vaccinated with rabies vaccine (RabV) 5 doses. In the study of post-exposure immune program, the protective rate of LipoRabV 3 was higher than that of control RabV5. The immunization procedure of lyophilized human rabies liposome vaccine was preliminarily determined: the immunization procedure before exposure was 0 ~ 14 days after exposure, 1 dose of freeze-dried human rabies liposome vaccine (6.0IU/) was inoculated before exposure, and after exposure, the procedure of immunization was 0 ~ 3 ~ 3 ~ 14 days each. 1 dose of lyophilized human rabies liposome vaccine (6.0IU/). Compared with the existing vaccine, the procedure of immunization before exposure was reduced from 3 to 2, and the whole immunization was reduced from 28 days to 14 days. After exposure, the immunization procedure was reduced from 5 to 3, and the whole immunization was reduced from 28 to 14 days.
