大鼠慢性牙周炎合并動(dòng)脈粥樣硬化動(dòng)物模型的建立及白細(xì)胞介素-6和C反應(yīng)蛋白表達(dá)分析

【摘要】: 目的:用Wistar大鼠建立動(dòng)脈粥樣硬化(atherosclerosis,As)合并慢性牙周炎(chronic periodontitis,CP)動(dòng)物模型,以探討冠心病與慢性牙周炎可能的相關(guān)性。方法:60只大鼠隨機(jī)分為四組,每組15只:A正常對(duì)照組,B慢性牙周炎組,C動(dòng)脈粥樣硬化組,D慢性牙周炎合并動(dòng)脈粥樣硬化組。各組接受相應(yīng)的建模處理。實(shí)驗(yàn)周期為12周。每20天檢查牙周指標(biāo)并測(cè)定體質(zhì)量。建模結(jié)束后,檢測(cè)血脂水平及ELISA法檢測(cè)血清白介素6(interleukin-6,IL-6)、C反應(yīng)蛋白(C-reactive protein,CRP)水平。組織病理學(xué)觀察各組實(shí)驗(yàn)牙牙周組織及動(dòng)脈血管病理改變。結(jié)果:牙周檢查發(fā)現(xiàn),B、D組附著喪失水平(attachment loss,AL)、齦溝出血指數(shù)(sulcular bleeding index,SBI)較A、C組有明顯增加,且差別有統(tǒng)計(jì)學(xué)意義(P0.05)。C、D組TC、TG、LDL水平較A、B組明顯升高(P0.05),D組血清IL-6、CRP水平均高于其他組(P0.05)。組織病理檢查發(fā)現(xiàn)B組動(dòng)脈血管中膜可見不均勻增厚。C、D組主動(dòng)脈及胸、腹主動(dòng)脈形成動(dòng)脈粥樣硬化病變。結(jié)論:結(jié)合牙周檢查、血脂檢測(cè)和組織病理學(xué)檢查綜合判定,初步成功建立大鼠CP合并As動(dòng)物模型。對(duì)各組IL-6、CRP水平的分析,CP可能通過炎癥因子的作用影響As的發(fā)生發(fā)展。
[Abstract]:Aim: to establish an animal model of atherosclerosis (atherosclerosis,As) with chronic periodontitis (chronic periodontitis,CP) in Wistar rats and to explore the possible correlation between coronary heart disease and chronic periodontitis. Methods: sixty rats were randomly divided into four groups, 15 rats in each group: a control group, B chronic periodontitis group, C atherosclerosis group, D chronic periodontitis combined with atherosclerosis group. Each group was treated with corresponding modeling. The experimental period was 12 weeks. Periodontal indexes were examined and body mass was measured every 20 days. After modeling, serum lipid level and serum interleukin 6 (interleukin-6,IL-6), C reactive protein) (C-reactive protein,CRP) level were detected by ELISA method. Histopathological changes of periodontal tissues and arterial vessels of experimental teeth were observed. Results: periodontal examination showed that the level of attachment loss (attachment loss,AL) and gingival sulcus bleeding index (sulcular bleeding index,SBI) in group B were significantly higher than those in group A (P 0.05), and the TC,TG,LDL level in group C was significantly higher than that in group A (P0.05). The level of serum IL-6,CRP in group C was significantly higher than that in group A (P0.05). Histopathological examination showed that there was an uneven thickening of the medial membrane of the arteries in group B. Atherosclerosis was found in the aorta, thoracic and abdominal aorta in group C and D. Conclusion: combined with periodontal examination, blood lipid detection and histopathological examination, the animal model of CP combined with As was established successfully. The level of IL-6,CRP in each group was analyzed. CP may influence the development of As through the action of inflammatory factors.
