EX527對(duì)熱量限制保護(hù)SH-SY5Y細(xì)胞的影響

【摘要】:目的觀察SIRT1抑制劑EX527對(duì)熱量限制處理的人神經(jīng)母細(xì)胞瘤株SH-SY5Y細(xì)胞的影響。方法體外培養(yǎng)SHSY5Y細(xì)胞,分為正常對(duì)照組(NC組)、熱量限制組(CR組)和熱量限制加SIRT1抑制劑組(CR+EX527組),光鏡下觀察細(xì)胞形態(tài)學(xué)改變,測(cè)定各組細(xì)胞噻唑藍(lán)(thiazolyl blue tetrazolium bromide,MTT)代謝率及乳酸脫氫酶(lactate dehydrogenase,LDH)漏出率,應(yīng)用蛋白免疫印跡(Western blotting)法測(cè)定SIRT1表達(dá)量。結(jié)果與NC組相比,CR組細(xì)胞形態(tài)好,表現(xiàn)為細(xì)胞胞體飽滿、突起伸展、貼壁牢固,MTT代謝率升高(P0.05),LDH漏出率有降低趨勢(shì),CR組和CR+EX527組SIRT1表達(dá)均增加(P0.05);與CR組相比,CR+EX527組細(xì)胞形態(tài)較差,突起回縮,貼壁性不好,MTT代謝率降低(P0.05),LDH漏出率有升高趨勢(shì),SIRT1表達(dá)略減少。結(jié)論熱量限制對(duì)神經(jīng)細(xì)胞具有保護(hù)作用,而SIRT1抑制劑EX527能夠減弱熱量限制的神經(jīng)保護(hù)作用,提示熱量限制對(duì)SH-SY5Y細(xì)胞的保護(hù)作用可能是由SIRT1介導(dǎo)的。
[Abstract]:Objective to observe the effect of SIRT1 inhibitor EX527 on human neuroblastoma cell line SH-SY5Y treated with heat restriction.Methods SHSY5Y cells were cultured in vitro and divided into normal control group (NC group), caloric restriction group (CR group) and caloric restriction plus SIRT1 inhibitor group (CR EX527 group). Morphological changes were observed under light microscope.The metabolic rate of thiazolyl blue tetrazolium bromidede (MTT) and the leakage rate of lactate dehydrogenase (LDH) were measured. The expression of SIRT1 was detected by Western blotting.Results compared with NC group, the cell morphology of CR group was better than that of NC group, which showed that the cell body was plump, the protuberance was extended, the metabolic rate of adherent to the cell wall was increased, the leakage rate of SIRT1 was decreased in CR group and CR EX527 group, the expression of SIRT1 in CR group and CR EX527 group was higher than that in CR group, and the cell morphology in CR EX527 group was worse than that in CR group.Protruding back, MTT metabolic rate decreased, the leakage rate of LDH increased and the expression of SIRT1 decreased slightly.Conclusion caloric restriction has protective effect on nerve cells, while SIRT1 inhibitor EX527 can attenuate the neuroprotective effect of caloric restriction on SH-SY5Y cells, suggesting that the protective effect of caloric restriction on SH-SY5Y cells may be mediated by SIRT1.
