單因素刺激方法致大鼠腎精虧虛的動(dòng)物模型及腎精虧虛對(duì)免疫功能影響的相關(guān)研究

【摘要】: 研究目的:通過研究單因素刺激方法致大鼠腎精虧虛的動(dòng)物模型,探討腎精虧虛對(duì)免疫功能影響,為臨床感染性疾病的防治起一定的指導(dǎo)作用。 研究方法:基于“房勞傷腎”的理論,采用人工單因素刺激致腎精虧虛的方法,在持續(xù)的一段時(shí)間內(nèi),通過自制電刺激儀連續(xù)刺激大鼠的敏感部位使其生殖器充血勃起并排出精液,建立單因素刺激所致腎精虧虛的動(dòng)物模型;造模結(jié)束后,分析天平測(cè)定大鼠睪丸濕重,HE染色法觀察睪丸組織形態(tài)變化,使用放射免疫法測(cè)定大鼠血清睪酮的含量;全血細(xì)胞分析儀測(cè)定大鼠血液中紅細(xì)胞和血紅蛋白含量;分析天平測(cè)定大鼠胸腺和脾臟的濕重,使用酶聯(lián)免疫法檢測(cè)大鼠血清IgG濃度和IL-2濃度;感染肺炎克雷伯菌后,使用酶聯(lián)免疫法檢測(cè)大鼠血清TNF-a濃度和IL-6濃度。 實(shí)驗(yàn)結(jié)果: (1)造模后睪丸的體積縮小,濕重顯著減輕。 (2)睪丸組織呈萎縮狀態(tài):大部分曲細(xì)精管退化變性,管腔內(nèi)生殖上皮變薄,各級(jí)精母細(xì)胞減少排列稀疏,精子減少甚至沒有精子,間質(zhì)水腫,曲細(xì)精管之間結(jié)締組織增多,界膜增厚并纖維化透明樣變性。對(duì)照組大鼠各級(jí)精母細(xì)胞排列有序,精子增多,界膜變薄,間質(zhì)水腫減輕。 (3)血清睪酮的含量模型一組較正常組降低,模型二組明顯高于正常組,左歸丸對(duì)照組高于模型一組但低于模型二組,右歸丸對(duì)照組均低于模型一組和模型二組。 (4)血液紅細(xì)胞和血紅蛋白含量模型一組與正常組相當(dāng),模型二組的血液紅細(xì)胞和血紅蛋白低于正常組,左歸丸對(duì)照組和右歸丸對(duì)照組高于模型二組。 (5)血清IgG濃度模型一組升高,模型二組最高,左歸丸對(duì)照組較模型一組升高但低于模型二組,右歸丸對(duì)照組均低于模型一組和模型二組。 (6)IL-2含量模型一組高于正常組,模型二組較正常組降低,左歸丸對(duì)照組和右歸丸對(duì)照組均高于模型一組和模型二組。 (7)經(jīng)肺炎克雷伯菌感染后,血清TNF-a濃度正常感染組和正常組相當(dāng),模型一感染組的血清TNF-a濃度均較正常感染組降低;血清TNF-a濃度左歸丸對(duì)照感染組接近正常感染組,右歸丸對(duì)照感染組較正常感染組升高。 (8)經(jīng)肺炎克雷伯菌感染后,血清IL-6含量正常感染組較正常組升高,模型一感染組的血清IL-6含量較正常感染組降低,左歸丸對(duì)照組和右歸丸對(duì)照組的血清IL-6含量均低于正常感染組。 研究結(jié)論: (1)通過人工單因素刺激的方法可以建立大鼠的腎精虧虛動(dòng)物模型。 (2)人工單因素刺激的方法致腎精虧虛可以導(dǎo)致機(jī)體免疫功能紊亂。 (3)人工單因素刺激的方法致腎精虧虛存在容易感染疾病的趨勢(shì)。
[Abstract]:Objective: to investigate the effect of deficiency of kidney essence on immune function in rats with deficiency of kidney essence by single factor stimulation, and to play a guiding role in the prevention and treatment of clinical infectious diseases. Methods: based on the theory of "kidney injury caused by room strain", the method of artificial single factor stimulation was used to induce deficiency of kidney essence, which lasted for a period of time. The animal model of kidney essence deficiency caused by single factor stimulation was established by continuous stimulation of sensitive parts of the genitals of rats with congestive erection and ejection of semen by self-made electric stimulator, and the model was made after the completion of the model. The morphological changes of testis were observed by HE staining method of wet weight of testis, the content of testosterone in serum was measured by radioimmunoassay, the contents of erythrocytes and hemoglobin in blood of rats were measured by whole blood cell analyzer. The wet weight of thymus and spleen of rats was measured by balance, serum IgG and IL-2 were detected by enzyme linked immunosorbent assay (Elisa), and serum TNF-a and IL-6 were detected by Elisa after infection with Klebsiella pneumoniae (Klebsiella pneumoniae). Experimental results:. 1) the volume of testis was reduced and the wet weight was significantly reduced after modeling. (2) testicular tissue showed atrophy: most of the seminiferous tubules degenerated and denatured, the reproductive epithelium thinned in the lumen, the number of spermatocytes decreased and sparse, the spermatozoa decreased or even did not have sperm, the interstitial edema, and the connective tissue between seminiferous tubules increased. In the control group, the spermatocytes arranged orderly, the sperm increased, the boundary membrane became thinner, and the interstitial edema was alleviated. The serum testosterone content in model group 1 was lower than that in normal group, the level of serum testosterone in model group 2 was significantly higher than that in normal group, the level of serum testosterone in model group 1 was higher than that in model group 1 but lower than that in model group 2, and that in control group of Yougui pill was lower than that in model group 1 and model 2. The contents of erythrocyte and hemoglobin in the model group were similar to those in the normal group. The erythrocyte and hemoglobin in the model group 2 were lower than those in the normal group, and the contents of red blood cell and hemoglobin in the model group were higher than those in the control group and the right Gui pill group. Serum IgG concentration in model group 1 was higher than that in model group 2, the level of serum IgG in control group was higher than that in model group 1 but lower than that in model group 2, and that in control group was lower than that in model group 1 and model 2. The level of IL-2 in model group 1 was higher than that in normal group, and the level of IL-2 in model group 2 was lower than that in normal group. The level of IL-2 in model group 1 and group 2 were higher than those in group 1 and group 2. (7) after infection with Klebsiella pneumoniae, the serum TNF-a concentration in the normal infection group was similar to that in the normal group, the serum TNF-a concentration in the model first infection group was lower than that in the normal infection group, and the serum TNF-a concentration in the control group was close to that in the normal infection group. The control group of Yougui pills was higher than the normal group. After infection with Klebsiella pneumoniae, the serum IL-6 content in the normal infection group was higher than that in the normal group, and the serum IL-6 content in the model first infection group was lower than that in the normal infection group. The levels of serum IL-6 in the control group and Yougui pill group were lower than those in the normal infection group. The study concluded that:. 1) the animal model of deficiency of kidney essence can be established by artificial single factor stimulation. 2) deficiency of kidney essence caused by artificial single factor stimulation can lead to disorder of immune function. 3) deficiency of kidney essence caused by artificial single factor stimulation has the tendency of infection easily.
