【摘要】: 樹突狀細(xì)胞(Dendritic cells,DC)是目前已知的功能最強(qiáng)的抗原遞呈細(xì)胞(APC),參與抗原的識(shí)別、加工處理與遞呈,是機(jī)體免疫反應(yīng)的首要環(huán)節(jié),也是免疫反應(yīng)中識(shí)別自我與非我的關(guān)鍵環(huán)節(jié)。近年來(lái)國(guó)際上對(duì)DC的研究進(jìn)展很快,對(duì)于DC在腫瘤免疫、感染免疫、移植免疫、自身免疫等方面作用的認(rèn)識(shí)不斷深入。但是由于DC僅占外周血單個(gè)核細(xì)胞總數(shù)的0.5%~1%,難以分離純化;而且常規(guī)的基礎(chǔ)DC培養(yǎng)基中主要成分為粒細(xì)胞-巨噬細(xì)胞集落刺激因子(GM-CSF),其雖然能促進(jìn)DC細(xì)胞的成熟,但在體外大量培養(yǎng)和擴(kuò)增以達(dá)到治療用途,尚存在一定問(wèn)題。這限制了DC的臨床應(yīng)用。因此,如何獲得大量高純度的成熟DC是DC研究的方向之一。 蛋氨酸腦啡肽(Methionine enkephalin ,MEK)對(duì)自然殺傷細(xì)胞、單核細(xì)胞、巨噬細(xì)胞和細(xì)胞因子的釋放都有調(diào)節(jié)作用,而且MEK能夠增強(qiáng)自然殺傷細(xì)胞活性和誘導(dǎo)白細(xì)胞介素2、白細(xì)胞介素4的分泌。因此,本課題嘗試用MEK作為共刺激因子,研究其對(duì)人外周血來(lái)源的DC細(xì)胞分化成熟的作用,探討MEK在體外刺激DC細(xì)胞,促進(jìn)其成熟的可行性。
[Abstract]:Dendritic cells (DC) is the most powerful antigen presenting cell known at present. It is involved in antigen recognition, processing and presentation. It is the most important link of immune response, and also the key link of self-recognition and non-self recognition in immune response.In recent years, there has been a rapid development in the research of DC in the world. The role of DC in tumor immunity, infection immunity, transplantation immunity, autoimmunity and so on has been deeply understood.However, since DC only accounts for 0.5% of the total number of peripheral blood mononuclear cells, it is difficult to isolate and purify, and the main component of conventional basic DC culture medium is granulocyte-macrophage colony stimulating factor (GM-CSFN), although it can promote the maturation of DC cells.However, in vitro culture and amplification to achieve therapeutic purposes, there are still some problems.This limits the clinical application of DC.Therefore, how to obtain a large number of high-purity mature DC is one of the research directions.Methionine enkephalin can regulate the release of natural killer cells, monocytes, macrophages and cytokines, and MEK can enhance the activity of natural killer cells and induce the secretion of interleukin 2 and interleukin 4.Therefore, we try to use MEK as a costimulatory factor to study the effect of MEK on the differentiation and maturation of DC cells from human peripheral blood, and to explore the feasibility of stimulating DC cells by MEK in vitro and promoting their maturation.
