【摘要】：目的:本研究旨在探討臍血間充質(zhì)干細胞對T淋巴細胞亞群及其細胞因子分泌的影響。方法:采用密度梯度離心法分離培養(yǎng)擴增的臍血間充質(zhì)干細胞,應用流式細胞術檢測臍血間充質(zhì)干細胞與淋巴細胞混合培養(yǎng)后T淋巴細胞亞群及其細胞因子IL-4和IFN-γ的變化情況。結果:T淋巴細胞與臍血間充質(zhì)干細胞混合培養(yǎng)后,淋巴細胞增殖受到抑制,CD4+T細胞亞群增加,CD8+T細胞亞群減少;根據(jù)細胞因子的變化,Th1和Tc1比率較對照組有顯著降低,而Th2和Tc2的比率卻有輕度上升。結論:臍血間充質(zhì)干細胞能夠抑制T淋巴細胞尤其是CD8+T細胞的增殖。此外,它還能降低與T淋巴細胞共培養(yǎng)體系中的Th1和Tc1細胞,升高Th2和Tc2細胞。臍血MSCs具有臨床應用潛力,可望預防和治療GVHD。
[Abstract]:Objective: This study aimed to investigate the effect of umbilical cord blood mesenchymal stem cells on T lymphocyte subsets and cytokines. Methods: isolated cultured mesenchymal stem cells from umbilical cord blood by density gradient centrifugation and the detection of changes of umbilical cord blood mesenchymal stem cells mixed with lymph cell culture after T lymphocyte subsets and cytokines IL-4 and IFN- by using flow cytometry. Results: T cells and human umbilical cord blood mesenchymal stem cells after co cultured lymphocytes proliferation was inhibited, CD4+T cell subsets increased, decreased CD8+T cell subsets; cytokines according to the changes of Th1 and Tc1 ratio was significantly lower than the control group, and the ratio of Th2 and Tc2 has slightly increased. Conclusion: umbilical cord blood mesenchymal stem cells can inhibit T lymphocytes especially CD8+T cell proliferation. In addition, it can also reduce and T lymphocytes co cultured Th1 and Tc1 cells, The increase of Th2 and Tc2 cells. The umbilical cord blood MSCs has potential clinical application to prevention and treatment of GVHD.
