體外擴(kuò)增巨核細(xì)胞的研究

【摘要】: 目的:探討采用臍血單個(gè)核細(xì)胞為起始細(xì)胞在無血清培養(yǎng)基中體外擴(kuò)增巨核細(xì)胞;觀察胎兒肺部組織來源的間充質(zhì)干細(xì)胞對臍血單個(gè)核細(xì)胞誘導(dǎo)分化為巨核細(xì)胞的影響。方法:分取臍血單個(gè)核細(xì)胞,在TPO,IL—11,肝素的擴(kuò)增體系中分為兩組培養(yǎng),第一組以CD34~+細(xì)胞作為對照,單獨(dú)培養(yǎng),第二組與間充質(zhì)干細(xì)胞共培養(yǎng),在7天,10天,14天時(shí),取細(xì)胞進(jìn)行活細(xì)胞計(jì)數(shù),流式細(xì)胞儀分析CD41a和CD61的表達(dá)情況。取14天細(xì)胞采用免疫組化和透射電鏡分析細(xì)胞形態(tài)與結(jié)構(gòu),通過流式細(xì)胞儀分析細(xì)胞DNA含量。結(jié)果:MNC經(jīng)細(xì)胞因子誘導(dǎo)后,CD41a和CD61陽性比例明顯上升,與0天時(shí)相比,培養(yǎng)14天后,CD41a~+細(xì)胞平均擴(kuò)增了429.5倍,CD61~+細(xì)胞平均擴(kuò)增了339倍。第二組在10天時(shí)得到的CD41~+細(xì)胞和CD61~+細(xì)胞最多,分別是第一組的4.5倍和4.7倍,免疫組化和電鏡結(jié)果表明,兩組細(xì)胞核發(fā)育不成熟。結(jié)論:在TPO,IL—11,肝素的擴(kuò)增體系下以MNC為起始細(xì)胞能夠誘導(dǎo)得到大量的巨核細(xì)胞,胎肺間充質(zhì)干細(xì)胞能有效刺激CD41~+細(xì)胞和CD61~+細(xì)胞生成,對細(xì)胞核發(fā)育影響。
[Abstract]:Objective: to investigate the expansion of megakaryocytes from cord blood mononuclear cells in serum-free medium in vitro. To observe the effect of mesenchymal stem cells derived from fetal lung tissue on the differentiation of cord blood mononuclear cells into megakaryocyte. The amplification system of heparin was divided into two groups. The first group was cultured alone with CD34 ~ cells and the second group co-cultured with mesenchymal stem cells. The living cells were counted and the expression of CD41a and CD61 were analyzed by flow cytometry. The morphology and structure of the cells were analyzed by immunohistochemistry and transmission electron microscopy on the 14th day. Results the positive rates of CD41a and CD61 were significantly increased after induced by cytokines. Compared with 0 days, the positive rates of CD41a and CD61 were significantly increased after 14 days of culture. CD41a~ cells were amplified by an average of 429.5 times and CD61- cells by 339 times on average. In the second group, the number of CD41 ~ cells and CD61 ~ cells were the highest at 10 days. The results of immunohistochemistry and electron microscope showed that the nuclei of the two groups were immature. In the system of heparin amplification, a large number of megakaryocytes could be induced by using MNC as the starting cells, while fetal lung mesenchymal stem cells could effectively stimulate the generation of CD41 ~ cells and CD61 ~ cells. Effect on nuclear development.
