【摘要】: 心理應(yīng)激是指有機(jī)體在某種環(huán)境刺激作用下由于客觀要求和應(yīng)付能力不平衡所產(chǎn)生的一種適應(yīng)環(huán)境的緊張反應(yīng)狀態(tài)。在現(xiàn)代社會(huì)中,各種各樣的應(yīng)激原如激烈的競(jìng)爭(zhēng)、快速的工作節(jié)奏、逐漸增大的生活壓力等等,都在影響著人們的生理和心理狀態(tài)。短期或適度的心理應(yīng)激可提高機(jī)體的警惕水平,有益于人體的健康和功能活動(dòng),但心理應(yīng)激狀態(tài)時(shí)間過(guò)長(zhǎng)或強(qiáng)度過(guò)大就會(huì)對(duì)人體產(chǎn)生有害作用。如今,心理應(yīng)激與心身疾病發(fā)生發(fā)展的關(guān)系已經(jīng)越來(lái)越受到人們的重視?谇徽5囊Ш瞎δ苁蔷捉兰 D下頜關(guān)節(jié)、牙牙合關(guān)系在中樞神經(jīng)系統(tǒng)作用下相互支持,相互制約的結(jié)果,而咬肌是口頜面部重要的咀嚼肌之一,它參與了口腔咀嚼、吞咽、言語(yǔ)等多種功能活動(dòng)。另外,升頜肌中肌梭等本體感受器分布密度高于降頜肌,而咬肌又是升頜肌中肌梭分布密度最高、對(duì)各種應(yīng)激狀態(tài)的反應(yīng)最為及時(shí)、敏感的一對(duì)肌肉,F(xiàn)代醫(yī)學(xué)針對(duì)心理應(yīng)激對(duì)口頜肌影響的研究已取得了一定的進(jìn)展。臨床流行病學(xué)調(diào)查證實(shí)心理應(yīng)激與口頜系統(tǒng)疾病之間存在密切關(guān)系,本系列研究的前期動(dòng)物實(shí)驗(yàn)也證明,咬肌的形態(tài)學(xué)、肌組織代謝、肌纖維牙型轉(zhuǎn)化、肌肉疼痛閾等諸多方面都與心理因素密不可分。然而,心理應(yīng)激對(duì)咬肌影響的機(jī)制仍不明確,還有待于進(jìn)一步深入的研究。 因此,本實(shí)驗(yàn)在課題組前期工作的基礎(chǔ)上,選取咬肌作為切入點(diǎn),采用心理應(yīng)激交流箱建立大鼠心理應(yīng)激模型,在檢測(cè)動(dòng)物行為學(xué)改變以確保實(shí)驗(yàn)大鼠處于心理應(yīng)激狀態(tài)的基礎(chǔ)上,觀察咬肌組織中自由基代謝、HSP70表達(dá)的變化,并計(jì)算二者之間的相關(guān)性,為更深入研究心理應(yīng)激導(dǎo)致口頜肌疲勞、功能紊亂乃至口頜系統(tǒng)疾病的機(jī)制提供實(shí)驗(yàn)依據(jù),并為口頜肌功能紊亂的防治奠定實(shí)驗(yàn)基礎(chǔ)。 本實(shí)驗(yàn)共分為三個(gè)部分。第一部分實(shí)驗(yàn)觀察大鼠心理應(yīng)激后的行為學(xué)變化。選取72只雄性SD大鼠,隨機(jī)分為心理應(yīng)激組(Psychological stress group, PS組)、足部電擊組(Footshock group,FS組)和空白對(duì)照組,每組24只。各組再按照實(shí)驗(yàn)時(shí)間分為1周、3周和5周三個(gè)亞組,其中空白對(duì)照組不接受任何外界刺激,FS組大鼠接受足部電擊,僅作為應(yīng)激原,最后將PS組和對(duì)照組大鼠納入實(shí)驗(yàn)研究。整個(gè)實(shí)驗(yàn)期間所有大鼠同條件飼養(yǎng)。在各時(shí)間點(diǎn),測(cè)量大鼠體重,采用高架十字迷宮(Elevated plus maze,EPM)實(shí)驗(yàn)和動(dòng)物曠場(chǎng)(Open field)實(shí)驗(yàn)觀察心理應(yīng)激對(duì)大鼠行為學(xué)特征的影響。EPM實(shí)驗(yàn)結(jié)果顯示:隨著心理應(yīng)激時(shí)間的增長(zhǎng),PS組大鼠進(jìn)入開(kāi)臂的次數(shù)逐漸減少,呈現(xiàn)時(shí)間依賴(lài)性,進(jìn)入開(kāi)臂的時(shí)間在心理應(yīng)激3周和5周后與對(duì)照組比較出現(xiàn)差異,5周時(shí)達(dá)到最低;3周和5周PS組大鼠進(jìn)入閉臂次數(shù)少于對(duì)照組,而各實(shí)驗(yàn)時(shí)間點(diǎn)在閉臂停留的時(shí)間卻均明顯增長(zhǎng);并且后兩個(gè)時(shí)間點(diǎn)PS組大鼠在EPM中的移動(dòng)距離也較對(duì)照組減少。曠場(chǎng)實(shí)驗(yàn)結(jié)果顯示:PS組大鼠應(yīng)激3周和5周后,曠場(chǎng)移動(dòng)速度和移動(dòng)距離小于對(duì)照組,中央?yún)^(qū)活動(dòng)時(shí)間隨心理應(yīng)激時(shí)間遞減,且均低于對(duì)照組。 第一部分實(shí)驗(yàn)證實(shí)了心理應(yīng)激可以影響大鼠的行為,在此基礎(chǔ)上我們進(jìn)行第二部分實(shí)驗(yàn)。大鼠心理應(yīng)激建模第1、3、5周行為學(xué)觀察后處死大鼠,取咬肌組織檢測(cè)自由基代謝情況,結(jié)果顯示:肌組織中SOD活力和GSH-PX活力在3周和5周應(yīng)激組中出現(xiàn)降低,但5周PS組較3周PS組的兩種抗氧化酶酶活性均有回升;CAT活力3周和5周PS組均低于同時(shí)間點(diǎn)對(duì)照組;PS組大鼠咬肌中MDA含量在3周時(shí)最多,到5周時(shí)又開(kāi)始減少,但仍高于對(duì)照組;H2O2含量?jī)H3周PS組與對(duì)照組有差異。 隨后,在第三部分實(shí)驗(yàn)中,我們運(yùn)用免疫組織化學(xué)染色和Western blot方法檢測(cè)咬肌中HSP70的表達(dá)情況。結(jié)果發(fā)現(xiàn):1周和3周PS組大鼠咬肌中熱休克蛋白70(HSP70)表達(dá)無(wú)明顯變化,而心理應(yīng)激5周后,大鼠的HSP70表達(dá)較同時(shí)間點(diǎn)對(duì)照組增強(qiáng),亦高于1周和3周PS組大鼠。同時(shí),我們計(jì)算HSP70相對(duì)灰度值與自由基代謝之間的相關(guān)性,發(fā)現(xiàn)肌組織中HSP70的表達(dá)與MDA含量呈負(fù)相關(guān)。 實(shí)驗(yàn)結(jié)論: 1、心理應(yīng)激交流箱可有效建立大鼠心理應(yīng)激模型;心理應(yīng)激可以引起大鼠焦慮樣行為。 2、心理應(yīng)激狀態(tài)能夠引起大鼠咬肌自由基代謝異常,這可能是心理應(yīng)激導(dǎo)致咀嚼肌功能紊亂的機(jī)制之一。 3、長(zhǎng)期心理應(yīng)激引起大鼠咬肌中HSP70的表達(dá)增強(qiáng),而HSP70的表達(dá)增強(qiáng)可能控制自由基代謝,減輕自由基對(duì)咀嚼肌的損傷。
[Abstract]:Psychological stress refers to an environment adapting to environmental stress caused by imbalance of objective requirement and coping ability when an organism is stimulated by some environment. In modern society, all kinds of stressors, such as fierce competition, fast pace of work and increasing pressure of life, are all affecting people's physiological and psychological state. Short or moderate psychological stress can improve the vigilance level of the body, and is beneficial to the health and function activities of the human body. But when the psychological stress is too long or the intensity is too large, it will have a harmful effect on the human body. Nowadays, the relationship between psychological stress and the development of psychosomatic disease has been paid more and more attention. Occlusal function is normal oral masticatory muscles, teeth of temporomandibular joint, the relationships of mutual support in the role of the central nervous system, the result of mutual restriction, and orofacial masseter muscle is an important one of the masticatory muscles, it is involved in the function of chewing, swallowing, oral speech and other activities. In addition, the distribution density of proprioceptor in the ascending mandibular muscle is higher than that in the descending jaw muscle, and masseter muscle is the most timely and sensitive muscle in the ascending jaw muscle with the highest density of muscle spindle and the response to various stress states. Modern medicine has made some progress in the study of the influence of psychological stress on the oral maxillary muscle. Clinical epidemiological investigation confirmed that there is a close relationship between psychological stress and stomatognathic system disease, previous animal experiments in this study also proved that the masseter muscle tissue morphology and metabolism of muscle fiber, tooth type conversion, muscle pain threshold and other aspects are inseparable and psychological factors. However, the mechanism of the effect of psychological stress on the masseter muscle is still unclear, and it remains to be further studied.
