依托泊苷誘導(dǎo)蛋白2.4調(diào)控iNKT細(xì)胞的發(fā)育和IFN-γ應(yīng)答(英文)

iNKT細(xì)胞是一類特殊的固有樣T淋巴細(xì)胞,在感染、腫瘤、自身性免疫疾病和代謝類疾病中都發(fā)揮重要的調(diào)控作用.揭示調(diào)控iNKT細(xì)胞發(fā)育、分化和功能的細(xì)胞分子機制,對于闡釋iNKT細(xì)胞與疾病的關(guān)系以及尋求可能的治療途徑都具有重要的意義.依托泊苷誘導(dǎo)蛋白2.4 (Etoposide-induced protein 2.4,Ei24)可調(diào)控細(xì)胞生長、凋亡和自噬等多種生物學(xué)功能,但其對iNKT細(xì)胞的發(fā)育和功能的影響仍不清楚.本研究利用Cre/loxP重組酶系統(tǒng)成功構(gòu)建T細(xì)胞中Ei24特異性敲除小鼠.敲除Ei24后,iNKT細(xì)胞在胸腺中的發(fā)育受到明顯抑制,肝臟和脾臟等組織中的iNKT比例和數(shù)目明顯減少.進一步研究發(fā)現(xiàn),Ei24主要影響iNKT1和iNKT17 2個亞群,對iNKT2的調(diào)控作用相對較小.當(dāng)腹腔注射iNKT細(xì)胞特異性活化抗原α-GC后,敲除Ei24后iNKT細(xì)胞的IFN-γ應(yīng)答更低,但不影響i NKT細(xì)胞的IL-4應(yīng)答.以上結(jié)果表明,Ei24可以調(diào)控iNKT細(xì)胞的發(fā)育與功能.
