剪應(yīng)力誘導(dǎo)骨髓間充質(zhì)干細(xì)胞向內(nèi)皮細(xì)胞分化的研究

【摘要】: 目的:研究剪應(yīng)力對(duì)骨髓間充質(zhì)干細(xì)胞(MSCs)分化的作用。方法:設(shè)計(jì)制作平行平板流動(dòng)腔系統(tǒng),將人骨髓間充質(zhì)干細(xì)胞在3、8、15dyn/cm2剪應(yīng)力水平分別作用24h,觀察細(xì)胞形態(tài)變化,并進(jìn)行內(nèi)皮細(xì)胞標(biāo)記物VonWillebrandfactor(vWF)定性間接免疫熒光染色,進(jìn)行Dil標(biāo)記的乙;疞DL(Dil-Ac-LDL)攝取實(shí)驗(yàn)、管柱結(jié)構(gòu)形成實(shí)驗(yàn)以鑒定是否具有內(nèi)皮細(xì)胞功能。結(jié)果:制作的平行平板流動(dòng)腔腔體透明、無(wú)毒,流體通過(guò)可形成穩(wěn)定層流。MSCs在剪應(yīng)力作用24h后,形態(tài)變得偏圓、呈多角形,平行于流體方向排列。MSCs在接受3、8dyn/cm2剪應(yīng)力作用24h后vWF染色陽(yáng)性,提示內(nèi)皮細(xì)胞表型特征,同時(shí)Dil-Ac-LDL攝取實(shí)驗(yàn)陽(yáng)性、在基質(zhì)膠上可形成管柱結(jié)構(gòu),提示具有內(nèi)皮細(xì)胞功能。而接受15dyn/cm2剪應(yīng)力作用的MSCs,vWF染色與Dil-Ac-LDL攝取實(shí)驗(yàn)均陰性,提示未出現(xiàn)向內(nèi)皮細(xì)胞分化。結(jié)論:本實(shí)驗(yàn)制作的平行平板流動(dòng)腔可以作為產(chǎn)生剪應(yīng)力的體外細(xì)胞培養(yǎng)系統(tǒng)。剪應(yīng)力可誘導(dǎo)MSCs向內(nèi)皮細(xì)胞分化,并且與力的大小相關(guān)。
