乳酸桿菌培養(yǎng)上清抑制慢性酒精大鼠小腸上皮TLR4-TBK1通路

【摘要】:目的:探討保加利亞乳酸桿菌培養(yǎng)上清(supernatant recovered from lactobacillus bulgaricus culture MRS broth,LBG-S)對(duì)慢性酒精攝入大鼠小腸上皮細(xì)胞Toll樣受體4(Toll-like receptor 4,TLR4)-TANK結(jié)合激酶-1(TANK binding kinase-1,TBK1)信號(hào)通路的作用。方法:雄性健康Wistar大鼠30只(2月齡,體重250~300 g)分為三組:2月齡對(duì)照組(即基線對(duì)照組),順應(yīng)1周后即處死;9月齡對(duì)照組,自由取食標(biāo)準(zhǔn)鼠糧及飲用雙蒸水7月;慢性酒精組,9月齡,飲用含25%乙醇的雙蒸水連續(xù)6月。處死大鼠后,分離培養(yǎng)并計(jì)數(shù)各組大鼠小腸黏膜中的大腸桿菌和乳酸桿菌;分離并培養(yǎng)各組大鼠的小腸上皮細(xì)胞,在有或無LBG-S(10μg/m L)預(yù)處理的情況下,給予脂多糖(lipopolysaccharide,LPS,10 EU/m L)刺激后,Western blot檢測(cè)各組大鼠小腸分離上皮細(xì)胞中的TLR4及TBK1水平,酶聯(lián)免疫吸附法檢測(cè)各組分離小腸上皮細(xì)胞培養(yǎng)上清中的腫瘤壞死因子-α(tumor necrosis factor-α,TNF-α)和干擾素-γ(interferon-γ,IFN-γ)。結(jié)果:慢性飲酒后,大鼠腸腔內(nèi)乳酸桿菌數(shù)量明顯低于相應(yīng)對(duì)照組(P0.05);大腸桿菌數(shù)量無明顯增加。LPS能明顯升高各組分離大鼠小腸上皮細(xì)胞TLR4、TBK1以及生成TNF-α和IFN-γ的水平(P0.05),且慢性酒精組升高幅度明顯大于2月齡及9月齡對(duì)照組(P0.05)。LBG-S的預(yù)處理能明顯抑制LPS對(duì)各組分離大鼠小腸上皮細(xì)胞TLR4、TBK1以及生成TNF-α和IFN-γ水平的上調(diào)作用(P0.05)。結(jié)論:慢性酒精攝入導(dǎo)致大鼠小腸上皮TLR4-TBK1通路對(duì)LPS的高敏感性,LBG-S能明顯抑制這一高敏感性。
