間充質(zhì)干細(xì)胞分化為心肌細(xì)胞過(guò)程中細(xì)胞形態(tài)變化與Nkx2.5表達(dá)相關(guān)性的初步研究

【摘要】:體外BMSCs分化為心肌細(xì)胞過(guò)程中,細(xì)胞在形態(tài)及相關(guān)基因的表達(dá)上發(fā)生明顯的變化。本研究以心肌組織裂解液誘導(dǎo)BMSCs,觀察分化過(guò)程中細(xì)胞形態(tài)變化,根據(jù)心肌細(xì)胞發(fā)生過(guò)程中細(xì)胞形態(tài)變化規(guī)律,選定5個(gè)檢測(cè)點(diǎn),行PCR檢測(cè)細(xì)胞中Nkx2.5的表達(dá),以探究分化過(guò)程中細(xì)胞形態(tài)變化與相關(guān)基因表達(dá)規(guī)律及其之間的相關(guān)性。實(shí)驗(yàn)結(jié)果表明BMSCs分化為心肌細(xì)胞的過(guò)程中,細(xì)胞形態(tài)變化呈現(xiàn)明顯規(guī)律:由單個(gè)大核,胞漿飽滿,折光性好的紡錘形;逐漸演變?yōu)槎虠U狀;形成突起并連接成網(wǎng)狀結(jié)構(gòu);細(xì)胞端相接呈"竹節(jié)樣"改變;最后形成肌管樣結(jié)構(gòu);誘導(dǎo)前細(xì)胞不表達(dá)Nkx2.5,誘導(dǎo)過(guò)程中隨著細(xì)胞形態(tài)的不斷變化Nkx2.5持續(xù)表達(dá)并逐漸增強(qiáng)。說(shuō)明間充質(zhì)干細(xì)胞分化為心肌細(xì)胞的過(guò)程中,Nkx2.5表達(dá)相對(duì)量的變化與細(xì)胞的形態(tài)變化可能有一定的相關(guān)。
[Abstract]:During the differentiation of BMSCs into cardiomyocytes in vitro, there were obvious changes in morphology and expression of related genes. In this study, BMSCs, induced by myocardial lysate was used to observe the changes of cell morphology during differentiation. According to the changes of cell morphology during cardiomyogenesis, five detection points were selected and the expression of Nkx2.5 in the cells was detected by PCR. The purpose of this study was to explore the relationship between the morphological changes of cells and the expression of related genes in the process of differentiation. The results showed that during the process of differentiation of BMSCs into cardiomyocytes, the morphological changes of the cells showed obvious regularity: from a single nucleus, full cytoplasm and good refractive spindle, gradually evolved into short rod, formed protuberance and joined into reticular structure, and developed into a short rod, formed a protuberance and joined together into a reticular structure. The terminal phase of the cells was changed as "bamboo node-like", and finally the myotubule-like structure was formed. During the induction process, the cells did not express Nkx2.5, and the Nkx2.5 continued to express and gradually increased with the continuous changes of the cell morphology. These results suggest that the relative amount of Nkx2.5 expression may be related to the morphological changes of MSCs in the process of differentiation into cardiomyocytes.
