建立動(dòng)脈粥樣硬化穢濁痰阻證動(dòng)物模型的研究

【摘要】:目的:研究動(dòng)脈粥樣硬化穢濁痰阻證動(dòng)物模型的建立。方法將60只載脂蛋白E基因敲除小鼠隨機(jī)分為4組,即高脂飼料和干寒環(huán)境聯(lián)合干預(yù)的模型組、高脂飼料對(duì)照組、干寒環(huán)境對(duì)照組和空白對(duì)照組。各組小鼠分別施加寒冷干燥環(huán)境和高膽固醇飼料等相關(guān)干預(yù)。喂養(yǎng)至第12周觀察并記錄小鼠體重、行為、大小便、上臂耐受力、舌象等生物學(xué)表征;處死后采集血液并檢測(cè)血脂等生化指標(biāo);光學(xué)顯微鏡觀察小鼠主動(dòng)脈標(biāo)本的細(xì)胞形態(tài)學(xué)的變化。將相關(guān)指標(biāo)量化后各組之間進(jìn)行比較。結(jié)果1.經(jīng)過(guò)觀察分析,聯(lián)合干預(yù)高脂飼料和干寒環(huán)境的實(shí)驗(yàn)組小鼠所反映的穢濁痰阻證候表征最為突出;2.光鏡觀察主動(dòng)脈細(xì)胞形態(tài),各組小鼠動(dòng)脈均可觀察到粥樣斑塊和內(nèi)膜的增厚;經(jīng)過(guò)高膽固醇飼料和干寒環(huán)境對(duì)均內(nèi)膜/管腔比值的作用分析,兩種干預(yù)因素對(duì)內(nèi)膜/管腔比值單獨(dú)作用和交互作用的影響差異有統(tǒng)計(jì)學(xué)意義(P<0.05);即高脂飼料和干寒環(huán)境對(duì)內(nèi)膜增厚有促進(jìn)作用,使內(nèi)膜/管腔比值增高,而兩種因素同時(shí)聯(lián)合干預(yù)的模型組小鼠動(dòng)脈粥樣硬化病變更為突出。結(jié)論置于干燥寒冷環(huán)境下喂予高膽固醇飼料的模型組小鼠的生物學(xué)表征和實(shí)驗(yàn)室檢查結(jié)果表明此組小鼠的宏觀、微觀表現(xiàn)能夠模擬動(dòng)脈粥樣硬化穢濁痰阻證,說(shuō)明造模成功。
[Abstract]:Objective: to study the establishment of animal model of atherosclerotic turbid phlegm obstruction syndrome. Methods 60 apolipoprotein E knockout mice were randomly divided into 4 groups: model group with high fat diet and dry cold environment, high fat diet control group, dry cold environment control group and blank control group. Each group of mice were given cold and dry environment and high cholesterol diet and other related interventions. The body weight, behavior, defecation, upper arm tolerance and tongue image of the mice were observed and recorded at the 12th week of feeding, and the blood samples were collected and blood lipids were measured. The morphological changes of mouse aorta were observed by optical microscope. The relative indexes were compared after quantification. Result 1. Through observation and analysis, the experimental group mice with combined intervention of high fat diet and dry cold environment reflected the most prominent symptoms of the syndrome of turbidity and phlegm obstruction; 2. Atheromatous plaques and intimal thickening were observed in the arteries of each group. The effects of high cholesterol diet and dry cold environment on the ratio of mean intima to lumen were analyzed. There were significant differences between the two intervention factors on the ratio of intima to lumen (P < 0.05). That is to say, high fat diet and dry cold environment can promote intimal thickening and increase the ratio of intima to lumen, while the atherosclerotic lesion of the model group treated by two kinds of factors at the same time is more prominent. Conclusion the biological characteristics and laboratory results of the model group fed with high cholesterol diet in dry and cold environment show that the macroscopic and microscopic manifestations of this group can mimic the syndrome of atherosclerotic turbidity and phlegm obstruction, which indicates that the model is successful.
