【摘要】：目的對一株Ⅰ型單純皰疹病毒(HSV-1)進行分離鑒定和增值特性研究。方法從病人口唇皰液樣本中分離得到一株病毒,經噬斑純化、形態(tài)鑒定、序列分析,確定該分離株為HSV-1,被命名為HSVIZW6A。用其分別感染Vero細胞、KMB17細胞、A375細胞,觀察其細胞病變情況,并繪制生長曲線,分析病毒的生物學特性。結果 HSVI-ZW6A株病毒以MOI=10感染不同細胞,細胞外和細胞內病毒在0~4h,病毒復制緩慢,4~12h病毒增殖進入對數生長期,12~24h病毒增殖緩慢,進入平臺期,但24h之后病毒效價測定顯示病毒滴度開始呈現下降的趨勢,且細胞外病毒最大量高于細胞內,且HSVI-ZW6A株病毒毒力上高于HSVI-strain17病毒。結論通過本實驗成功分離和鑒定了一株Ⅰ型HSV病毒,并對其生物學特性進行了分析,為今后的實驗奠定了基礎。
[Abstract]:Objective to isolate and identify a herpes simplex virus (HSV-1) strain. Methods A virus was isolated from a sample of oral blister fluid from patients. After plaque purification, morphological identification and sequence analysis, it was confirmed that the isolate was named HSV-1, as HSVIZW6A.. Vero cells, KMB17 cells and A375 cells were infected respectively. The cytopathic changes of A375 cells were observed, and the growth curves were plotted to analyze the biological characteristics of the virus. Results the virus of HSVI-ZW6A strain was infected with different cells by MOI=10. The replication of the virus was slow at 0hs and within the cells, the proliferation of the virus at 412h entered the logarithmic growth phase, and the virus proliferated slowly at 1224 h, and entered the plateau phase. However, after 24 hours, the titer of the virus began to decrease, and the maximum amount of the extracellular virus was higher than that of the intracellular virus, and the virulence of the HSVI-ZW6A strain was higher than that of the HSVI-strain17 virus. Conclusion A strain of HSV virus type I was successfully isolated and identified by this experiment, and its biological characteristics were analyzed, which laid a foundation for future experiments.
