一株Ⅰ型單純皰疹病毒的分離鑒定及增殖特性

【摘要】:目的對(duì)一株Ⅰ型單純皰疹病毒(HSV-1)進(jìn)行分離鑒定和增值特性研究。方法從病人口唇皰液樣本中分離得到一株病毒,經(jīng)噬斑純化、形態(tài)鑒定、序列分析,確定該分離株為HSV-1,被命名為HSVIZW6A。用其分別感染Vero細(xì)胞、KMB17細(xì)胞、A375細(xì)胞,觀察其細(xì)胞病變情況,并繪制生長曲線,分析病毒的生物學(xué)特性。結(jié)果 HSVI-ZW6A株病毒以MOI=10感染不同細(xì)胞,細(xì)胞外和細(xì)胞內(nèi)病毒在0~4h,病毒復(fù)制緩慢,4~12h病毒增殖進(jìn)入對(duì)數(shù)生長期,12~24h病毒增殖緩慢,進(jìn)入平臺(tái)期,但24h之后病毒效價(jià)測(cè)定顯示病毒滴度開始呈現(xiàn)下降的趨勢(shì),且細(xì)胞外病毒最大量高于細(xì)胞內(nèi),且HSVI-ZW6A株病毒毒力上高于HSVI-strain17病毒。結(jié)論通過本實(shí)驗(yàn)成功分離和鑒定了一株Ⅰ型HSV病毒,并對(duì)其生物學(xué)特性進(jìn)行了分析,為今后的實(shí)驗(yàn)奠定了基礎(chǔ)。
[Abstract]:Objective to isolate and identify a herpes simplex virus (HSV-1) strain. Methods A virus was isolated from a sample of oral blister fluid from patients. After plaque purification, morphological identification and sequence analysis, it was confirmed that the isolate was named HSV-1, as HSVIZW6A.. Vero cells, KMB17 cells and A375 cells were infected respectively. The cytopathic changes of A375 cells were observed, and the growth curves were plotted to analyze the biological characteristics of the virus. Results the virus of HSVI-ZW6A strain was infected with different cells by MOI=10. The replication of the virus was slow at 0hs and within the cells, the proliferation of the virus at 412h entered the logarithmic growth phase, and the virus proliferated slowly at 1224 h, and entered the plateau phase. However, after 24 hours, the titer of the virus began to decrease, and the maximum amount of the extracellular virus was higher than that of the intracellular virus, and the virulence of the HSVI-ZW6A strain was higher than that of the HSVI-strain17 virus. Conclusion A strain of HSV virus type I was successfully isolated and identified by this experiment, and its biological characteristics were analyzed, which laid a foundation for future experiments.
