【摘要】：目的:在原核系統(tǒng)內(nèi)獲得高表達(dá)的人血小板衍生生長(zhǎng)因子BB(PGDF-BB),并對(duì)形成的包涵體進(jìn)行復(fù)性。方法:對(duì)PGDF-BB核酸編碼序列進(jìn)行優(yōu)化,構(gòu)建pET-22b-PGDF-BB表達(dá)載體,以提高PCDF-BB的表達(dá)量;優(yōu)化PGDF-BB包涵體復(fù)性條件,提高蛋白復(fù)性率和生物活性。結(jié)果:構(gòu)建了pET-22b-PGDF-BB高效表達(dá)載體,原核表達(dá)的重組人PGDF-BB占細(xì)菌總蛋白的25%,PGDF-BB包涵體復(fù)性率達(dá)到15%。結(jié)論:對(duì)表達(dá)序列的優(yōu)化設(shè)計(jì)可顯著提高蛋白的表達(dá)量,復(fù)性方法的改良提高了蛋白的復(fù)性率和生物活性。
[Abstract]:Aim: to obtain highly expressed human platelet-derived growth factor BB (PGDF-BB) in prokaryotic system and renaturation of the inclusion bodies. Methods: the PGDF-BB nucleic acid coding sequence was optimized to construct the pET-22b-PGDF-BB expression vector to increase the expression of PCDF-BB, to optimize the PGDF-BB inclusion body renaturation conditions and to improve the protein renaturation rate and biological activity. Results: the high efficient expression vector of pET-22b-PGDF-BB was constructed. The recombinant human PGDF-BB expressed in prokaryotic expression accounted for 25% of the total bacterial protein. The inclusion body renaturation rate of PGDF-BB reached 15%. Conclusion: the optimized design of expression sequence can significantly increase the expression of protein, and the improvement of renaturation method can improve the renaturation rate and biological activity of protein.
