孕期亞臨床甲減大鼠模型的建立

【摘要】:目的建立可靠的孕期亞臨床甲減大鼠動(dòng)物模型。方法將60只成年雌性Wistar大鼠隨機(jī)分為三組:1完全甲減組:手術(shù)完全切除甲狀腺;2亞臨床甲減組:手術(shù)完全切除甲狀腺后,予L-T4微滲泵1.0μg/(100 g·d)濃度持續(xù)皮下泵入,自懷孕第15 d劑量改為1.05μg/(100 g·d);3正常對(duì)照組:行甲狀腺假手術(shù)。各組大鼠在基礎(chǔ)狀態(tài)、孕前和懷孕后第10、13、17 d和分娩當(dāng)天采血測(cè)定甲狀腺功能。結(jié)果各組大鼠在基礎(chǔ)狀態(tài)時(shí),血清TSH和TT4水平差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。完全甲減組大鼠在孕前和整個(gè)懷孕過(guò)程中及分娩當(dāng)天血清TSH水平始終明顯高于亞臨床甲減組和正常對(duì)照組(P均0.01),血清TT4水平始終明顯低于其他兩組(P均0.01)。亞臨床甲減組大鼠在孕前和整個(gè)懷孕過(guò)程中及分娩當(dāng)天血清TSH水平始終明顯高于正常對(duì)照組(P均0.05),血清TT4水平始終與正常對(duì)照組相當(dāng)(P均0.05)。結(jié)論應(yīng)用手術(shù)完全切除甲狀腺,再予微滲泵皮下持續(xù)輸入合適劑量的L-T4可以成功建立孕期亞臨床甲減大鼠模型。
[Abstract]:Objective to establish a reliable animal model of subclinical hypothyroidism during pregnancy. Methods 60 adult female Wistar rats were randomly divided into three groups: 1 complete hypothyroidism group: complete thyroidectomy; 2Subclinical hypothyroidism group: after total thyroidectomy, L-T4 microosmotic pump 1.0 渭 g / (100 g d) was continuously injected subcutaneously, and the dose was changed to 1.05 渭 g / (100 g d);) from the 15th day of pregnancy. 3 normal control group: thyroid sham-operation was performed. The thyroid function was measured at the basal state, at 1317 days before pregnancy and on the 10th day of pregnancy and on the day of parturition. Results there was no significant difference in serum TSH and TT4 levels between the three groups (P0.05). The serum TSH level in the complete hypothyroidism group was significantly higher than that in the subclinical hypothyroidism group and the normal control group before pregnancy and during pregnancy and on the day of delivery (P0.01), and the serum TT4 level was significantly lower than that in the other two groups (P0.01). The serum TSH levels in the subclinical hypothyroidism group were significantly higher than those in the normal control group before pregnancy, during pregnancy and on the day of delivery. The serum TT4 level was always similar to that of the normal control group (P0. 05). Conclusion the model of subclinical hypothyroidism in pregnant rats can be successfully established by surgical resection of thyroid gland and subcutaneous continuous infusion of appropriate dose of L-T4 with microosmotic pump.
