日本血吸蟲SjC23-Hsp70DNA疫苗與IL-12對水牛保護(hù)性作用的研究

【摘要】: 目的:研究日本血吸蟲中國大陸株23 kDa膜蛋白-熱休克蛋白(SjC23-Hsp70)DNA疫苗聯(lián)合佐劑白細(xì)胞介素-12(IL-12)質(zhì)粒DNA對水牛的免疫保護(hù)作用。 方法:將購自血吸蟲病非流行區(qū)8-10月齡健康水牛45頭隨機(jī)分為A組(SjC23-Hsp70+IL-12,300μg)、B組(SjC23+IL-12,300μg)和C組(pVAX+IL-12,300μg)等3組,每組15頭。每頭牛經(jīng)肩部肌注免疫3次,每次間隔為28天。末次免疫后28天,每頭牛感染日本血吸蟲尾蚴1000條。解剖前2天及當(dāng)天分別收集糞便1次,用定量法檢測蟲卵和毛蚴數(shù)。攻擊感染后56天解剖所有水牛,經(jīng)胸主動脈灌沖法收集成蟲,計(jì)數(shù)成蟲數(shù),檢測每克肝組織蟲卵數(shù)。試驗(yàn)采用雙盲法,所用疫苗及制劑均在實(shí)驗(yàn)結(jié)束后解碼。 結(jié)果:A、B組與C組相比,分別獲得45.70%和26.61%的減雌率,44.51%和25.84%的減蟲率,41.10%和31.63%的減糞卵率,48.11%和38.07%的減毛蚴率及43.39%和31.95%的減肝卵率。A組的5個(gè)率均比B組高。 結(jié)論:用SjC23-Hsp70+DNA疫苗和IL-12聯(lián)合免疫水?色@得明顯的免疫保護(hù)作用。
[Abstract]:Aim: to study the protective effect of 23 kDa membrane protein-heat shock protein (SjC23-Hsp70) DNA vaccine combined with adjuvant interleukin-12 (IL-12) plasmid DNA on buffalo. Methods: 45 healthy buffalo were randomly divided into three groups: group A (SjC23-Hsp70 IL-12300 渭 g), B (SjC23 IL-12300 渭 g) and group C (pVAX IL-12300 渭 g). Each cow was immunized with shoulder intramuscular injection for 3 times, with 28 days interval. 28 days after the last immunization, each cow was infected with 1000 cercariae of Schistosoma japonicum. The feces were collected 2 days before dissection and once on the same day, and the eggs and the number of cercariae were detected by quantitative method. All buffaloes were dissected 56 days after attack. Adult worms were collected by thoracic aortic perfusion. The number of adult worms was counted and the number of eggs per gram of liver tissue was detected. The double-blind method was used and the vaccine and preparation were decoded after the experiment. Results: 45.70% and 26.61% female reduction rate, 44.51% and 25.84% worm reduction rate, 41.10% and 31.63% fecal egg reduction rate were obtained in group A B and C, respectively. 48.11% and 38.07% of the cases and 43.39% and 31.95% of the liver oocytes were reduced respectively. The five rates in group A were higher than those in group B. Conclusion: buffalo immunized with SjC23-Hsp70 DNA vaccine and IL-12 can obtain obvious immune protective effect.
