雌馬酚對血管內(nèi)皮細胞凋亡和增殖影響

【摘要】:目的探討大豆異黃酮(SI)代謝物雌馬酚對血管內(nèi)皮細胞凋亡和增殖的影響。方法體外培養(yǎng)血管內(nèi)皮細胞(ECV-304),采用不同濃度雌馬酚(0、6.25、12.50、25.00、50.00和100.00μmol/L)處理24 h,收集細胞,采用流式細胞術(shù)分析細胞內(nèi)活性氧(ROS)、細胞凋亡和細胞周期;采用免疫組織化學技術(shù)分析凋亡相關蛋白Bax、Bcl-2和caspase-3表達;采用噻唑藍(MTT)法分析雌馬酚對ECV-304細胞增殖影響。結(jié)果與對照組比較,6.25μmol/L雌馬酚組ECV-304細胞內(nèi)ROS含量下降,100.00μmol/L雌馬酚組ECV-304細胞內(nèi)ROS含量升高;與對照組比較,25.00、50.00、100.00μmol/L雌馬酚組ECV-304細胞早、晚期凋亡率明顯升高,并可導致細胞G1期阻滯(P0.05);MTT實驗結(jié)果顯示,0、6.25、12.50、25.00、50.00、100.00μmol/L雌馬酚組ECV-304細胞增殖抑制率分別為-(8.23±2.21)%、-(3.29±1.07)%、(6.75±1.73)%、(25.46±4.82)%和(36.93±4.66)%,低劑量雌馬酚對細胞生長具有促進作用,高劑量雌馬酚對細胞生長具有抑制作用;與對照組比較,25.00μmol/L雌馬酚組作用24 h后,細胞內(nèi)Bax和caspase-3表達增加,而Bcl-2表達減少。結(jié)論低濃度雌馬酚可降低細胞內(nèi)ROS含量,促進ECV-304細胞增殖;高濃度雌馬酚可導致細胞凋亡、細胞周期阻滯,抑制細胞增殖。
[Abstract]:Objective to investigate the effect of estradiol, a metabolite of soybean isoflavone (SI), on apoptosis and proliferation of vascular endothelial cells. Methods Vascular endothelial cells (ECV-304) were cultured in vitro and treated with different concentrations of estradiol (0.6.25A12.5012.50,25.0050.00 and 100.00 渭 mol/L) for 24 hours. The cells were collected and analyzed by flow cytometry for intracellular reactive oxygen species (ROS),). Apoptosis and cell cycle; The expression of apoptosis-related protein Bax,Bcl-2 and caspase-3 was analyzed by immunohistochemistry and the effect of estradiol on the proliferation of ECV-304 cells was analyzed by thiazolyl (MTT) assay. Results compared with the control group, the content of ROS in ECV-304 cells in 6.25 渭 mol/L female equisol group decreased, and the ROS content in ECV-304 cells increased in 100.00 渭 mol/L estralophen group. Compared with the control group, the ECV-304 cells in the 25.00 渭 mol/L 50.00100.00 渭 mol/L group were early, the late apoptosis rate was significantly increased, and the G1 phase arrest was induced (P0.05). The results of MTT test showed that the inhibition rate of ECV-304 cell proliferation was-(8.23 鹵2.21)%, (3.29 鹵1.07)%, (6.75 鹵1.73)% and (6.75 鹵1.73)%, respectively. (25.46 鹵4.82)% and (36.93 鹵4.66)% respectively. Low dose of estradiol promoted cell growth, and high dose of female equisol inhibited cell growth. Compared with the control group, the expression of Bax and caspase-3 increased and the expression of Bcl-2 decreased in 25.00 渭 mol/L female equallol group for 24 h. Conclusion low concentration of estradiol can reduce the content of ROS and promote the proliferation of ECV-304 cells, and high concentration of estradiol can induce cell apoptosis, arrest cell cycle and inhibit cell proliferation.
