【摘要】：目的:探討肝素結(jié)合表皮生長(zhǎng)因子樣生長(zhǎng)因子(HB-EGF)對(duì)小鼠胚胎黏附和擴(kuò)展中的作用機(jī)制。方法:建立小鼠胚胎與小鼠子宮內(nèi)膜細(xì)胞共培養(yǎng)模型,分A、B、C、D、E共5組,分別加入不同濃度的HB-EGF(0μg/L、20μg/L、50μg/L、100μg/L、200μg/L),于倒置顯微鏡下統(tǒng)計(jì)各組胚胎黏附率及擴(kuò)展率;用免疫組織化學(xué)法檢測(cè)小鼠子宮內(nèi)膜細(xì)胞骨橋蛋白(OPN)的表達(dá)及定位,Western blotting法測(cè)定各濃度HB-EGF對(duì)子宮內(nèi)膜細(xì)胞OPN表達(dá)水平的影響,并分析其與HB-EGF的關(guān)聯(lián)。結(jié)果:OPN在小鼠子宮內(nèi)膜細(xì)胞高表達(dá),主要分布于胞質(zhì)中,隨HB-EGF濃度升高,小鼠子宮內(nèi)膜細(xì)胞中OPN的表達(dá)增強(qiáng),小鼠胚胎對(duì)子宮內(nèi)膜的黏附和擴(kuò)展率也逐漸增高。結(jié)論:HBEGF可能通過上調(diào)小鼠子宮內(nèi)膜細(xì)胞OPN的表達(dá),促進(jìn)小鼠胚胎對(duì)子宮內(nèi)膜的黏附和擴(kuò)展。
[Abstract]:Aim: to investigate the mechanism of heparin binding epidermal growth factor like growth factor (HB-EGF) on mouse embryo adhesion and expansion. Methods: a co-culture model of mouse embryo and mouse endometrial cells was established. The mice were divided into 5 groups, and were treated with different concentrations of HB-EGF (0 渭 g / L, 20 渭 g / L, 50 渭 g / L, 100 渭 g / L, 200 渭 g / L), respectively. The rates of embryo adhesion and expansion were calculated under inverted microscope. The expression of osteopontin (OPN) in mouse endometrial cells was detected by immunohistochemical method, and the effect of HB-EGF concentration on OPN expression in endometrial cells was determined by localization, Western blotting method, and its association with HB-EGF was analyzed. Results: the expression of OPN was highly expressed in mouse endometrial cells, mainly distributed in the cytoplasm. With the increase of HB-EGF concentration, the expression of OPN in mouse endometrial cells increased, and the adhesion and expansion rate of mouse embryos to endometrium increased gradually. Conclusion: HBEGF may promote the adhesion and expansion of mouse embryo to endometrium by up-regulating the expression of OPN in mouse endometrial cells.
