酒精攝入導(dǎo)致雌鼠及孕鼠子代血清泌乳素升高及其機(jī)制的探討

【摘要】:第一部分酒精對(duì)雌鼠血清泌乳素水平及垂體濕重的影響 目的探討酒精對(duì)雌鼠血清泌乳素(PRL)水平、垂體濕重的影響,了解攝取酒精的濃度和年齡與血清PRL水平改變及垂體增生的相關(guān)性,了解飲酒與高泌乳素血癥(HP)的關(guān)系。 方法1.40只SD同齡成年雌鼠隨機(jī)分為4組,每組分別予10%,20%,30%酒精灌胃4周和8周各5只,對(duì)照組予等容積和時(shí)間蒸餾水灌胃;2.30只4周、6周、8周齡的SD雌鼠分別予以20%酒精灌胃4周,各自對(duì)照組均予等容積蒸餾水灌胃;3.各組大鼠處死后均用放射免疫分析法(RIA)檢測(cè)血清PRL水平,稱垂體濕重,分析攝取酒精的濃度和年齡與血清PRL水平、垂體濕重的相關(guān)性。 結(jié)果1.灌胃4周時(shí),30%酒精組大鼠血清PRL水平和垂體濕重明顯高于其它三組;灌胃8周時(shí),10%和20%酒精組明顯高于同期30%酒精組和對(duì)照組。組內(nèi)比較,灌胃8周時(shí),10%及20%酒精組大鼠PRL水平和垂體濕重均顯著高于灌胃4周時(shí),30%酒精組大鼠PRL水平和垂體濕重顯著低于4周時(shí),以上各差異均有統(tǒng)計(jì)學(xué)意義,P0.05。2.同濃度酒精灌胃4周后,各年齡組與相應(yīng)的對(duì)照組比較,4周齡大鼠血清PRL水平和垂體濕重顯著升高;6周齡和8周齡大鼠血清PRL水平、垂體濕重差異均無(wú)統(tǒng)計(jì)學(xué)意義,P0.05。 結(jié)論1.酒精攝入可引起雌鼠血清PRL水平發(fā)生改變,且有劑量依賴性和累積效應(yīng):高濃度酒精可在較短時(shí)間內(nèi)使血清PRL水平升高,長(zhǎng)期低濃度酒精攝取可引起PRL水平顯著性增高。2.長(zhǎng)期大劑量酒精攝入可導(dǎo)致雌性大鼠垂體產(chǎn)生PRL的功能減退。3.較之成年期,幼年期雌性大鼠攝入酒精更易導(dǎo)致血清PRL水平升高。 第二部分?jǐn)z入酒精對(duì)孕鼠子代血清泌乳素及垂體濕重的影響 目的探討大鼠妊娠期大量攝入酒精對(duì)子代血清泌乳素水平、垂體濕重的影響。 方法6只孕鼠隨機(jī)分為2組,酒精組于懷孕第1天(P1)起給予20%酒精(2g/kg·d)灌胃至分娩前。對(duì)照組同期給予等量蒸餾水。子鼠喂養(yǎng)至4周后每組隨機(jī)抽取一半處死,余下的繼續(xù)喂養(yǎng)至8周處死。各組大鼠處死后均用放免法檢測(cè)血清泌乳素水平,并稱垂體濕重。 結(jié)果1.4周齡子鼠:酒精組的垂體濕重明顯高于對(duì)照組(P0.05),而兩組間PRL水平差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);2.8周齡子鼠:酒精組的PRL水平和垂體濕重均較對(duì)照組顯著升高(P0.05)。 結(jié)論大鼠妊娠期大量攝入酒精可使子代血清泌乳素水平升高、垂體濕重增加。酒精可能通過(guò)刺激垂體增生,使得血泌乳素水平升高。 第三部分酒精攝入導(dǎo)致雌鼠血清泌乳素升高機(jī)制的探討 目的:探討酒精導(dǎo)致雌鼠血清泌乳素升高的機(jī)制。 方法:40只大鼠隨機(jī)分為2組,實(shí)驗(yàn)組給予20%酒精2m1灌胃,每日2次共8周,對(duì)照組給予等體積蒸餾水灌胃。檢測(cè)血清泌乳素水平、垂體濕重及免疫組化觀察垂體腺細(xì)胞增殖情況,庫(kù)倫陣列電化學(xué)高效液相色譜法檢測(cè)下丘腦勻漿去甲腎上腺素、多巴胺、5-羥色胺、r-氨基丁酸、谷氨酸水平并進(jìn)行比較。 結(jié)果:1.實(shí)驗(yàn)組血清泌乳素水平、垂體濕重均顯著高于對(duì)照組,P0.05。2.實(shí)驗(yàn)組下丘腦勻漿多巴胺、5-羥色胺、r-氨基丁酸顯著低于對(duì)照組,谷氨酸濃度顯著高于對(duì)照組, P0.05。去甲腎上腺素濃度與對(duì)照組比較無(wú)顯著性差異,P0.05。3.實(shí)驗(yàn)組垂體細(xì)胞Ki-67的表達(dá)顯著高于對(duì)照組,P0.05。 結(jié)論:酒精可以導(dǎo)致泌乳素升高,其機(jī)制可能為:1.垂體催乳細(xì)胞增殖;2.酒精刺激引起下丘腦泌乳素釋放抑制因子(PIF)如多巴胺、5-羥色胺、r-氨基丁酸等分泌減少,而泌乳素釋放因子(PRF)如谷氨酸分泌增多。
