骨髓間充質(zhì)干細胞的磁標(biāo)記成像及移植治療骨缺血壞死的研究
[Abstract]:Avascular necrosis of the femoral head is a common clinical disease. The treatment of avascular necrosis of femoral head is based on the improvement of peripheral blood circulation except surgery. Bone marrow mesenchymal stem cells (BMSCs) are easy to take, can be expanded in vitro, and have a variety of differentiation potential. There is no immune rejection in transplantation of cells derived from autologous bone marrow, and the immunogenicity of allogeneic mesenchymal stem cells is very low. It provides a new seed cell for tissue damage repair. In this study, rMSCs was isolated and amplified by density gradient centrifugation. The cells were identified as bone marrow mesenchymal stem cells (BMSCs) by morphology and osteogenesis. The labeled rMSCs was labeled with phenidomagnetic poly-lysine and the labeling rate was 100. The in vitro imaging of labeled cells with 1.5TMRI showed that the GRE T2*WI sequence could be the preferred sequence in the study of magnetically labeled cells. A rabbit model of steroid-induced avascular necrosis of femoral head was established by injecting horse serum and methylprednisolone. Bone marrow mesenchymal stem cells (BMSCs) were transplanted into rabbit model of avascular necrosis of femoral head by interventional catheter technique. On this basis, fetal fMSCs was isolated and amplified by whole bone marrow culture method, and fMSCs magnetic labeling imaging was performed. The results were similar to those of rabbit MSCs. In this study, we obtained experience from rabbit bone marrow mesenchymal stem cell research and applied it to human bone marrow mesenchymal stem cell research step by step, hoping to make some useful exploration for the further clinical application of bone marrow mesenchymal stem cell transplantation in the treatment of avascular necrosis of femoral head.
