HPLC分析植物乳桿菌膽鹽水解酶降解特性

【摘要】:本文建立了同時檢測6種膽鹽的HPLC方法,并對兩株植物乳桿菌來源的膽鹽水解酶BSH-1降解特性進行了分析。HPLC色譜條件如下:Sepax Bio-C18(3μm,4.6 mm×150 mm);流速:0.6 m L/min;檢測波長:205 nm;流動相A:水/TFA=100/0.1,流動相B:乙腈/TFA=100/0.09;洗脫梯度如下:0 min~8 min,B 25%~35%;8 min~30 min,B 35%~48%;30 min~30.5 min,B48%~25%;30.5 min~40 min,B 25%。結(jié)果表明,本文建立的HPLC法可以快速準(zhǔn)確的測定BSH-1對膽鹽的降解活力及偏好,有利于篩選具有高效降解膽鹽的乳酸菌。兩種植物乳桿菌來源的BSH-1對GC、GCDC、GDC這三種膽鹽可以進行降解,尤其是GDC的降解率高達99.9%,但是對TAC、TCDC、TDC幾乎不能降解,也即是來源于植物乳桿菌的BSH-1對降解甘氨系列膽酸鹽的能力是高于;窍盗心懰猁}。
[Abstract]:In this paper, a HPLC method for simultaneous detection of 6 kinds of bile salts was established, and the degradation characteristics of bile salt hydrolase BSH-1 from two strains of Lactobacillus plantarum were analyzed. The chromatographic conditions of HPLC were as follows: Sepax Bio-C18 (3 渭 m) 4.6 mm 脳 150 mm); The flow rate was 0. 6 mL / min, the detection wavelength was 205 nm; mobile phase A: water / TFA=100/0.1, mobile phase B: acetonitrile / TFA=100/0.09; elution gradient was as follows: 0 min~8 min,B 25 + 35 + 8 min~30 min,B 35. 30 min~30.5 min,B48%~25%;30.5 min~40 min,B 25. The results showed that the HPLC method established in this paper could quickly and accurately determine the degradation activity and preference of BSH-1 to bile salt, and was beneficial to the screening of lactic acid bacteria with high degradation of bile salt. BSH-1 from two strains of Lactobacillus plantarum can degrade GC,GCDC,GDC, especially the degradation rate of GDC as high as 99.9%, but it can hardly degrade TAC,TCDC,TDC. The ability of BSH-1 derived from Lactobacillus plantarum to degrade glargine series cholate is higher than that of taurocholate series.
