【摘要】：目的探討西羅莫司(SRL)對小鼠異位心臟移植模型的移植物存活時間以及脾臟中調(diào)節(jié)性T細胞(Treg)分化和增殖的影響。方法應(yīng)用Cuff法建立雄性BALB/c→C57BL/6小鼠頸部異位心臟移植模型。術(shù)后隨機分為3組各10只受體:對照組術(shù)后不予特殊藥物治療,SRL組術(shù)后1~14 d予SRL 10 mg/(kg·d)灌胃,環(huán)孢素(Cs A)組術(shù)后1~14 d予Cs A 30 mg/(kg·d)灌胃。記錄移植心臟存活時間,于移植心停搏或術(shù)后14 d取脾,分離單個核細胞,上流式細胞儀檢測CD4+CD25+Treg占CD4+T細胞比例(CD4+CD25+Treg%),采用逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)法半定量檢測Foxp3信使核糖核酸(mRNA)表達水平。結(jié)果與對照組比較,Cs A組和SRL組均明顯延長小鼠移植心的生存時間(均為P0.01),而兩者之間比較差異無統(tǒng)計學意義(P0.05)。與對照組比較,Cs A組脾臟中CD4+CD25+Treg%明顯降低,而SRL組則明顯升高(均為P0.01),后兩組比較差異有統(tǒng)計學意義(P0.01)。SRL組脾臟T細胞的Foxp3 mRNA表達明顯高于對照組和Cs A組,其中對照組亦明顯高于Cs A組(均為P0.01)。結(jié)論在小鼠心臟移植模型中,SRL明顯延長移植物的存活期,促進CD4+CD25+Treg的增殖與生長,有利于免疫耐受的形成。
[Abstract]:Objective to investigate the effects of sirolimus (SRL) on graft survival time and (Treg) differentiation and proliferation of regulatory T cells in the spleen of mice with heterotopic heart transplantation. Methods the cervical heterotopic heart transplantation model of male BALB/c C57BL/6 mice was established by Cuff method. Each group was randomly divided into 3 groups: the control group received no special drug therapy, the SRL group received SRL 10 mg/ (kg d) intragastric perfusion on the 14th day after operation, and the cyclosporine (Cs A) group received Cs A30 mg/ (kg d) intragastric perfusion on the 1th day after operation. The survival time of the transplanted heart was recorded, and the spleen was taken 14 days after cardiac arrest, and the mononuclear cells were isolated. The ratio of CD4 CD25 Treg to CD4 T cells (CD4 CD25 Treg%) was detected by upflow cytometry, and the (mRNA) expression of Foxp3 messenger RNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results compared with the control group, Cs A group and SRL group significantly prolonged the survival time of heart transplantation in mice (P0.01), but there was no significant difference between the two groups (P0.05). Compared with the control group, CD4 CD25 Treg% in the spleen of the, Cs A group was significantly lower than that in the control group, while that in the SRL group was significantly higher than that in the control group (all P0.01). There was a significant difference between the latter two groups (P0.01). SRL group was significantly higher than the control group and Cs A group in Foxp3 mRNA expression of spleen T cells. The control group was also significantly higher than Cs A group (P0.01). Conclusion in mouse heart transplantation model, SRL significantly prolonged the survival period of the graft, promoted the proliferation and growth of CD4 CD25 Treg, and was beneficial to the formation of immune tolerance.
