【摘要】： 目的：構(gòu)建骨形態(tài)發(fā)生蛋白-2(BMP-2)真核表達(dá)質(zhì)粒pcDNA3.1-BMP-2,轉(zhuǎn)染兔骨髓間充質(zhì)干細(xì)胞(BMSCs)。檢測轉(zhuǎn)染后BMP-2基因的表達(dá)。 方法：(1)利用人外周血中抽提的基因組DNA,通過聚合酶鏈?zhǔn)椒磻?yīng)(PCR)合成BMP-2基因,將其與pcDNA3.1載體進(jìn)行基因重組,構(gòu)建表達(dá)質(zhì)粒pcDNA3.1-BMP-2。再通過PCR及DNA測序的方法檢測重組體。(2)利用pcDNA3.1-BMP-2轉(zhuǎn)染兔BMSCs,再通過RT-PCR、免疫組化和蛋白印跡法(Western blot)檢測BMP-2基因的表達(dá)。 結(jié)果：(1)經(jīng)PCR擴(kuò)增和DNA測序證實(shí),本實(shí)驗(yàn)構(gòu)建的重組質(zhì)粒目的基因片段為人BMP-2-cDNA。(2)經(jīng)RT-PCR、免疫組化和Western blot證實(shí),轉(zhuǎn)染pcDNA3.1-BMP-2后的兔BMSCs內(nèi)有大量BMP-2 mRNA的轉(zhuǎn)錄和蛋白的表達(dá),而未轉(zhuǎn)染的兔BMSCs未見陽性表達(dá)。 結(jié)論：(1)本實(shí)驗(yàn)成功構(gòu)建BMP-2真核表達(dá)質(zhì)粒并在兔BMSCs中得到表達(dá)。(2)本研究為進(jìn)一步研究用BMP-2基因轉(zhuǎn)染的方法來加速牽張成骨(Distraction osteogenesis DO)新骨形成的實(shí)驗(yàn)奠定了基礎(chǔ)。
[Abstract]:Objective: to construct eukaryotic expression plasmid pcDNA3.1-BMP-2, of bone morphogenetic protein-2 (BMP-2) and transfect (BMSCs). Into rabbit bone marrow mesenchymal stem cells (BMSCs). The expression of BMP-2 gene after transfection was detected. Methods: (1) the genomic DNA, extracted from human peripheral blood was used to synthesize BMP-2 gene by polymerase chain reaction (PCR). The BMP-2 gene was recombined with pcDNA3.1 vector to construct the expression plasmid pcDNA3.1-BMP-2.. Then the recombinant was detected by PCR and DNA sequencing. (2) the expression of BMP-2 gene was detected by RT-PCR, immunohistochemistry and Western blotting (Western blot). (2) Rabbit BMSCs, was transfected with pcDNA3.1-BMP-2. Results: (1) it was confirmed by PCR amplification and DNA sequencing that the target gene fragment of recombinant plasmid constructed in this study was human BMP-2-cDNA. (2) by RT-PCR, immunohistochemistry and Western blot, a large number of BMP-2 mRNA transcription and protein expression were found in the BMSCs transfected with pcDNA3.1-BMP-2. No positive expression was found in untransfected rabbit BMSCs. Conclusion: (1) the eukaryotic expression plasmid of BMP-2 was successfully constructed and expressed in rabbit BMSCs. (2) this study laid a foundation for further study on the method of BMP-2 gene transfection to accelerate the bone formation of distraction osteogenesis (Distraction osteogenesis DO).
