外周血調(diào)節(jié)性T淋巴細(xì)胞及Foxp3的表達(dá)與反復(fù)體外受精—胚胎移植失敗的相關(guān)性研究

【摘要】: 目的1探討外周血中CD4~+CD25~+調(diào)節(jié)性T細(xì)胞與反復(fù)體外受精-胚胎移植失敗之間的相關(guān)性;2探討外周血中Foxp3蛋白與mRNA的表達(dá)與反復(fù)體外受精-胚胎移植失敗之間的相關(guān)性。 方法收取原因不明反復(fù)體外受精-胚胎移植失敗者、正常非孕婦女及第一周期試管助孕成功者的外周靜脈血(肝素鈉抗凝)。用雙熒光標(biāo)記流式細(xì)胞分析技術(shù),檢測上述三組婦女外周血中CD4~+CD25~+T淋巴細(xì)胞及CD4~+CD25~(high)T淋巴細(xì)胞的表達(dá)情況;用免疫組化的方法檢測三組婦女外周血中Foxp3蛋白的表達(dá)情況;用RT-PCR的方法檢測三組婦女外周血中Foxp3 mRNA的表達(dá)。 結(jié)果1原因不明反復(fù)體外受精-胚胎移植失敗患者外周血中CD4~+CD25~+T淋巴細(xì)胞及CD4~+CD25~(high)T淋巴細(xì)胞的表達(dá)均低于正常非孕婦女和第一周期試管助孕成功者;第一周期試管助孕成功者外周血中CD4~+CD25~+T淋巴細(xì)胞的表達(dá)低于正常非孕婦女,但CD4~+CD25~(high)T淋巴細(xì)胞的表達(dá)與正常非孕婦女無統(tǒng)計(jì)學(xué)差異;2原因不明反復(fù)體外受精-胚胎移植失敗患者外周血中Foxp3蛋白的表達(dá)低于正常非孕婦女和第一周期試管助孕成功者,第一周期試管助孕成功者外周血中Foxp3蛋白的表達(dá)與正常非孕婦女無統(tǒng)計(jì)學(xué)差異;3原因不明反復(fù)體外受精-胚胎移植失敗患者外周血中Foxp3 mRNA的表達(dá)低于正常非孕婦女和第一周期試管助孕成功者,第一周期試管助孕成功者外周血中Foxp3 mRNA的表達(dá)與正常非孕婦女無統(tǒng)計(jì)學(xué)差異。 結(jié)論外周血CD4~+CD25~+調(diào)節(jié)性T細(xì)胞尤其是CD4~+CD25~(high)調(diào)節(jié)性T淋巴細(xì)胞及FOXP3的表達(dá)異?赡芘c原因不明反復(fù)體外受精-胚胎移植失敗的發(fā)生有相關(guān)性。
[Abstract]:Objective 1 To investigate the correlation between CD4 ~ + CD25 ~ + regulatory T cells and repeat in vitro fertilization-embryo transfer failure in peripheral blood, and to explore the correlation between the expression of Foxp3 protein and mRNA in peripheral blood and the failure of repeated in vitro fertilization-embryo transfer. Methods Peripheral venous blood (heparin) was collected from patients with unexplained recurrent in vitro fertilization-embryo transfer failure, normal non-pregnant women and first-cycle test tube assisted pregnancy. The expression of CD4 ~ + CD25 ~ + T lymphocytes and CD4 ~ + CD25 ~ (high) T lymphocytes in peripheral blood of three groups of women was detected by double fluorescence labeling flow cytometry. Expression of Foxp3 mRN in peripheral blood of three groups of women by RT-PCR Results 1 The expression of CD4 ~ + CD25 ~ + T lymphocytes and CD4 ~ + CD25 ~ (high) T lymphocytes in peripheral blood of patients with unexplained recurrent in vitro fertilization-embryo transplantation were lower than that of non-pregnant women and first. The expression of CD4 ~ + CD25 ~ + T lymphocytes in peripheral blood of the first cycle test tube was lower than that of non-pregnant women, but the expression of CD4 ~ + CD25 ~ (high) T lymphocytes was normal. The expression of Foxp3 protein in peripheral blood of patients with unexplained recurrent in vitro fertilization-embryo transfer failure was lower than that in non-pregnant women and the first cycle test tube, and the expression of Foxp3 protein in peripheral blood of the first cycle test tube was normal. The expression of Foxp3 mRNA in peripheral blood of patients with unexplained recurrent in vitro fertilization-embryo transfer failure was lower than that of non-pregnant women and the first cycle test tube, and the expression of Foxp3 mRNA in peripheral blood of the first cycle test tube was normal. Conclusion The abnormal expression of CD4 ~ + CD25 ~ + regulatory T cells, especially CD4 ~ + CD25 ~ (high) regulatory T lymphocytes and FOXP3 in peripheral blood may be associated with unexplained recurrent in vitro fertilization.
