膽石六號湯劑對豚鼠膽色素結石模型膽囊的抗炎作用

【摘要】:目的探究膽石六號湯劑對膽色素結石豚鼠膽囊腫瘤壞死因子-α(TNF-α)及核轉錄因子(NF-κB)表達的影響。方法將雌性豚鼠40只隨機分成3組,空白組10只,模型組15只,實驗組15只。空白組除外,其余2組均采用飲食誘導法,建立膽色素結石模型。實驗組,灌胃膽石六號湯劑7.7g·kg-1·d-1,其余2組均灌胃用同體積的0.9%氯化鈉。30 d后,觀察各組豚鼠成石率,用免疫組織化學技術檢測膽囊TNF-α及NF-κB蛋白表達和膽囊病理組織的改變情況。結果模型組成石率為90.62%,明顯高于空白組26.76%,表明造模成功;實驗組成石率為11.00%,明顯低于模型組90.62%,可見藥物具有抗膽結石的作用。與空白組比較,模型組的TNF-α、NF-κB蛋白表達量明顯增加(P0.01)。與模型組比較,實驗組的TNF-α、NF-κB蛋白表達量明顯較少(P0.05)。結論膽石六號湯劑可以明顯減少膽色素結石豚鼠的成石率,這可能與藥物降低膽色素結石豚鼠膽組織TNF-α、NF-κB蛋白的表達有關。
[Abstract]:Objective to investigate the effect of Ganshi No.6 decoction on the expression of tumor necrosis factor- 偽 (TNF- 偽) and nuclear transcription factor (NF- 魏 B) in gallbladder of pigmented gallstone guinea pigs. Methods 40 female guinea pigs were randomly divided into 3 groups: blank group (n = 10), model group (n = 15) and experimental group (n = 15). Except the blank group, the other two groups were induced by diet to establish the model of pigment gallstone. In the experimental group, 7. 7 g kg-1 d-1 of gavage gallstone no. 6 decoction, and 0. 9% sodium chloride in the other two groups were given intragastrically. 30 days later, the lithogenesis rate of guinea pigs in each group was observed, and the expression of TNF- 偽 and NF- 魏 B protein in gallbladder and the changes of gallbladder pathological tissue were detected by immunohistochemical technique. Results the rate of model stone composition was 90.62, which was significantly higher than that of blank group 26.76, which indicated that the model was successful, and the experimental composition stone rate was 11.00, which was significantly lower than that of model group 90.62. It can be seen that the drug has the effect of anti-gallstone. Compared with the blank group, the expression of TNF- 偽 and NF- 魏 B protein in the model group was significantly increased (P0.01). Compared with the model group, the expression of TNF- 偽 and NF- 魏 B protein in the experimental group was significantly lower (P0.05). Conclusion the gallstone formation rate of the guinea pigs with gallbladder pigment stone can be significantly reduced by the decoction. This may be related to the decrease of the expression of TNF- 偽 and NF- 魏 B protein in the gallbladder tissue of the guinea pigs with gallbladder pigment stone.
