正常雌性大鼠尿道橫紋括約肌肌球蛋白重鏈同源分子基因表達的研究

【摘要】:目的:尿道括約肌系統(tǒng)是哺乳類動物實現(xiàn)控尿的基本解剖結構,其中環(huán)繞尿道的橫紋括約肌是構成尿道括約肌系統(tǒng)的最重要結構成分。尿道括約肌系統(tǒng)的控尿功能表現(xiàn)得復雜而精細,以滿足動物在復雜多樣的日常活動中對控尿的需求。研究尿道橫紋括約肌纖維的類型和相對含量,可以更好地理解尿道橫紋括約肌發(fā)揮復雜而精細的控尿功能的生理機制。 方法:取正常成年未生育雌性Fisher 344大鼠12只,年齡12月,麻醉后解剖取出完整尿道,提取尿道總RNA,采用逆轉錄-聚合酶鏈反應(RT-PCR)法檢測尿道橫紋括約肌肌球蛋白重鏈同源分子mRNA的類型和相對含量,從而獲得尿道橫紋括約肌纖維類型及其相對含量的資料。 結果:正常成年未生育雌性大鼠尿道橫紋括約肌表達四種肌球蛋白重鏈同源分子mRNA,即MHC-Ⅰ、MHC-Ⅱa、MHC-Ⅱb和MHC-Ⅱx。其中MHC-Ⅱb的mRNA相對含量最高(38.1±0.7)%,MHC-Ⅱa次之(34.6±0.8)%,MHC-Ⅱx再次之(18.1±1.2)%,MHC-Ⅰ最低(9.2±0.4)%。 結論:正常成年未生育雌性大鼠尿道橫紋括約肌表達全部4種肌球蛋白重鏈同源分子,顯示尿道橫紋括約肌含有4種類型的纖維:typeⅠ(慢收縮纖維)、typeⅡa(快收縮抗疲勞纖維)、typeⅡb(快收縮易疲勞纖維)和typeⅡx(超快收縮纖維)。尿道橫紋括約肌多樣化的纖維類型是其實現(xiàn)復雜和精細控尿功能的分子基礎。
