【摘要】：肺炎嗜衣原體主要外膜蛋白是其特征抗原之一。實驗中通過PCR方法從肺炎嗜衣原體基因組中擴增主要外膜蛋白基因,插入pET32a(+)表達載體,轉(zhuǎn)化BL21(DE3)感受態(tài)細(xì)胞,得到表達56kD融合蛋白的工程菌株。該菌株的表達量可達53%,提純后的主要外膜蛋白純度可達90%以上,在Western Blotting試驗和膠體金免疫層析試驗中顯示了良好的抗原性。
[Abstract]:The main outer membrane protein of Chlamydia pneumoniae is one of its characteristic antigens. The main outer membrane protein gene was amplified from the genome of Chlamydia pneumoniae by PCR and inserted into pET32a () expression vector, and then transformed into BL21 (DE3) competent cells, and the engineering strain expressing 56kD fusion protein was obtained. After purification, the purity of the main outer membrane protein was over 90%, which showed good antigenicity in Western Blotting test and colloidal gold immunochromatographic assay.
