變性I型膠原對晚代骨髓間充質(zhì)細(xì)胞成骨能力的影響

【摘要】: 目的 探討變性的Ⅰ型膠原(DC)作為培養(yǎng)基質(zhì)對體外培養(yǎng)的骨髓間充質(zhì)干細(xì)胞的增殖和成骨能力的影響。 方法 體外培養(yǎng)大鼠的骨髓間充質(zhì)干細(xì)胞,分為A、B兩組,A組在鋪有變性的Ⅰ型膠原膜的培養(yǎng)皿中培養(yǎng);B組在沒有鋪?zhàn)冃缘蘑裥湍z原膜的培養(yǎng)皿中培養(yǎng)。培養(yǎng)至第八代時,分別檢測兩組細(xì)胞的增殖活性和細(xì)胞調(diào)亡率;并對細(xì)胞進(jìn)行成骨細(xì)胞誘導(dǎo),進(jìn)行堿性磷酸酶染色和定量分析。 結(jié)果 對于體外培養(yǎng)的第八代骨髓間充質(zhì)干細(xì)胞,相比較普通培養(yǎng)的細(xì)胞,生長在變性的Ⅰ型膠原膜上的細(xì)胞表現(xiàn)出較高的增殖活性和ALP表達(dá),并且有較少的細(xì)胞發(fā)生調(diào)亡。 結(jié)論 變性的Ⅰ型膠原作為骨髓間充質(zhì)干細(xì)胞的體外培養(yǎng)基質(zhì),可減緩細(xì)胞由于多次傳代而引起的細(xì)胞衰老,并可保持細(xì)胞的成骨分化能力。為組織工程提供了一種可獲得大量有增殖活性和分化能力的種子細(xì)胞的途徑。
