葡聚糖硫酸鈉在構建肝纖維化動物模型中的價值探討

【摘要】:目的探討葡聚糖硫酸鈉(DSS)對BALB/C小鼠肝損傷及其纖維化的影響。方法 50只BALB/C小鼠隨機分成正常組、對照組和觀察組(6、12周),每組10只,對照組小鼠用5%四氯化碳(CCl4)橄欖油溶液誘導肝纖維化模型,觀察組小鼠用3.5%DSS灌胃誘導肝損傷及纖維化模型。HE染色觀察肝臟病理損傷,Massion染色觀察肝臟纖維化情況,改良賴氏法檢測血清谷丙轉氨酶(ALT)和谷草轉氨酶(AST)活性,ELISA法檢測血清Ⅰ型前膠原羧基端原肽(PⅠCP)和Ⅲ型前膠原肽(PⅢNP)含量,免疫組化檢測肝臟α-平滑肌肌動蛋白(α-SMA)表達變化。結果對照組和觀察組小鼠各時間段肝臟的病理損傷明顯,膠原纖維表達量增多,血清中ALT和AST活性增強、PⅠNP和PⅢNP含量增加,肝內α-SMA表達量增加,上述變化以對照組最明顯,觀察組12周次之,觀察組與對照組比較,差異有統(tǒng)計學意義(P0.05)。結論 DSS能有效誘導BALB/C小鼠肝纖維化,其誘導機制值得深入研究。
