兔單側(cè)輸尿管可逆性不全梗阻模型制作及實(shí)驗(yàn)觀察

【摘要】: 目的:建立可逆性輸尿管部分梗阻(PUUO)動(dòng)物模型,并觀察部分梗阻及梗阻解除后腎臟功能及形態(tài)學(xué)改變。 方法:成年健康雄性大白兔40只,用縫合線在左側(cè)輸尿管近腎盂處半縫扎,建立PUUO動(dòng)物模型,分別于1、2、4、6、8月各隨機(jī)抽取8只,行超聲檢查,然后將其中4只取雙側(cè)腎臟送病檢,另外4只解除梗阻繼續(xù)飼養(yǎng),6個(gè)月后復(fù)查; 結(jié)果:梗阻后積水腎盂前后徑(APD)、腎盂容積(V)逐漸增大,最大腎實(shí)質(zhì)厚度(RCT)逐漸減小,與正常值比較有顯著性差異( P 0.01) ,各時(shí)間段之間比較有顯著性差異( P 0.01) ;梗阻解除后,APD、V逐漸變小,RCT逐漸增加,與梗阻解除前比較有顯著性差異( P 0.01) ,時(shí)間段之間比較有顯著性差異( P 0.01)。梗阻解除越早,恢復(fù)越快,病理學(xué)檢查也支持這一結(jié)果。 結(jié)論:該模型成功復(fù)制了PUUO所致腎臟形態(tài)學(xué)改變,是一種較理想的PUUO動(dòng)物模型。
[Abstract]:Objective: to establish a reversible ureteral obstruction (PUUO) animal model and observe the renal function and morphology after partial obstruction and obstruction.
Methods: 40 adult healthy male rabbits were sutured with suture on the left ureter near the renal pelvis. The PUUO animal model was established. 8 rats were randomly selected at 1,2,4,6,8 month, respectively, and 4 of them were taken from bilateral kidney for examination, and the other 4 were reared for 6 months after 6 months.
Results: after the obstruction of the hydronephrosis (APD), the volume of renal pelvis (V) increased gradually and the maximum renal parenchyma thickness (RCT) gradually decreased, and there was a significant difference compared with the normal value (P 0.01). There was a significant difference between each time period (P 0.01). After the obstruction was relieved, APD, V gradually became smaller, RCT gradually increased, and was more significant than before the removal of obstruction. Differences (P 0.01), there was a significant difference between the time periods (P 0.01). The earlier the obstruction was relieved, the quicker the recovery. Pathological examination also supported the result.
Conclusion: the model successfully replicated the renal morphological changes induced by PUUO, and was an ideal PUUO animal model.
