豚鼠與SD大鼠高脂血癥模型的比較研究
[Abstract]:Objective: to compare the characteristics of two animal hyperlipidemia models in guinea pig Sprague-Dawley (SD) rats and the effects of two kinds of animal hyperlipidemic serum on HUVEC injury, so as to provide experimental basis for selecting ideal animal models for the treatment and prevention of hyperlipidemia. Methods: 20 guinea pig SD rats were randomly divided into control group and model group. The control group was fed with basic diet, while the model group was fed with high fat diet. Blood samples were collected after 30 days of continuous feeding, and blood lipid indexes were measured. The liver and aorta were taken for pathological examination. Human umbilical vein endothelial cells (HUVEC) were cultured in each group. The levels of von Willebrand factor (vWF) in the supernatant were detected by Elisa. Results after 30 days of modeling, Compared with SD rat hyperlipidemia model group, the weight gain of guinea pigs in the guinea pig hyperlipidemia model group was more significant (P < 0.01) .2.The increase of LDL-C in guinea pigs was significantly higher than that in SD rats (P < 0.01). 3. The fatty degree of guinea pig liver was heavier than that of SD rats. (4) the activity of SOD in guinea pig liver tissue was significantly lower than that in SD rat (P < 0.05), and the arteriosclerosis index (AI) of guinea pig was significantly higher than that of SD rat (P < 0.01), while the activity of SOD in guinea pig liver was significantly lower than that in SD rat (P < 0.01), while the activity of SOD in guinea pig liver was lower than that in SD rat (P < 0.05). The intima of aorta could form atherosclerosis. 7, the damage degree of guinea-pig hyperlipidemia serum to HUEVC was more serious, and the content of vWF in guinea-pig supernatant was significantly higher than that in 24h (P < 0.01). Conclusion: the guinea pig is more suitable for the study of hyperlipidemia and as than the rats with anti-atherogenic ability.
