版納微型豬近交系動物瘢痕模型的建立(實驗Ⅱ)

【摘要】: [目的]以云南特有的版納微型豬近交系為對象,建立由動物自身產(chǎn)生的增生性瘢痕模型,為研究增生性瘢痕的發(fā)生機(jī)制及防治措施提供較為理想的動物模型。 [方法]于10只版納微型豬的10個背側(cè)制造切割傷創(chuàng)面(左右隨機(jī)):切除4cm×5cm的范圍的全層皮膚及肌筋膜層,每側(cè)2個,共計20個創(chuàng)面;另10個背側(cè)制造4cm×5cmⅢ°度燒傷創(chuàng)面,每側(cè)2個,共計20個創(chuàng)面。術(shù)后予80萬單位青霉素鈉注射劑肌注,每日兩次,連續(xù)三日,隔離飼養(yǎng)。 (1)術(shù)后連續(xù)12周觀察創(chuàng)面的自然愈合情況。 (2)對形成增生塊的創(chuàng)面進(jìn)行大體觀察、組織學(xué)檢測等并加入TGF-β_1及INF-γ觀察其對增生塊的作用。 [結(jié)果](1)切割傷創(chuàng)面均未見明顯增生塊形成,術(shù)后21天左右脫痂后創(chuàng)面于一周內(nèi)迅速縮小,6周后創(chuàng)面完全上皮化,有毛發(fā)長出。 (2)燒傷創(chuàng)面中90%發(fā)生形態(tài)與人增生性瘢痕類似的增生塊。 (3)燒傷創(chuàng)面增生塊組織學(xué)檢查見真皮中有大量成纖維細(xì)胞增殖并有少量膠原沉積,隨著時間推移,膠原成份增多,真皮層內(nèi)有膠原特征性的結(jié)節(jié)或漩渦狀結(jié)構(gòu)。 (4)局部注入TGF-β_1及INF-γ可促進(jìn)或抑制增生塊的發(fā)生。 [結(jié)論]版納微型豬近交系背側(cè)皮膚Ⅲ°燒傷后的創(chuàng)面經(jīng)自然愈合后可發(fā)生與人增生性瘢痕類似的病理改變,該模型是研究增生性瘢痕的發(fā)生機(jī)制及評估其治療方法的較好的動物模型之一。
