胚胎期大鼠體內(nèi)誘導(dǎo)免疫耐受應(yīng)用于造血干細(xì)胞移植的研究

【摘要】: 目的:通過(guò)大鼠胚胎期體內(nèi)誘導(dǎo)免疫耐受,預(yù)防GVHD,應(yīng)用于造血干細(xì)胞移植。為克服造血干細(xì)胞移植須MHC相合這一限制性障礙奠定實(shí)驗(yàn)基礎(chǔ)。 方法:①胚胎期供體卵黃囊內(nèi)接種受體MHC抗原。②受體大鼠經(jīng)Co608.0Gy照射后,行骨髓腔內(nèi)骨髓細(xì)胞移植。③移植后,觀察受體大鼠一般狀況,檢測(cè)受體大鼠血象變化,查骨髓象,用PCR法檢測(cè)受體外周血中供體來(lái)源的白細(xì)胞。④做GVHD相關(guān)病理檢查,并進(jìn)行皮膚移植。 結(jié)果:①實(shí)驗(yàn)組受體大鼠移植后100天成活率為86.7%,明顯高于各實(shí)驗(yàn)對(duì)照組,有明顯統(tǒng)計(jì)學(xué)差異。②實(shí)驗(yàn)組受體大鼠照射移植后,白細(xì)胞計(jì)數(shù)明顯下降,12天后回升,20天后升至正常水平;骨髓象示骨髓增生良好;用PCR法可檢測(cè)到供體來(lái)源的白細(xì)胞。以上示重建血液系統(tǒng)成功。③組織病理學(xué)檢查:實(shí)驗(yàn)組無(wú)GVHD現(xiàn)象,對(duì)照組有GVHD現(xiàn)象。④皮膚移植實(shí)驗(yàn)表明誘導(dǎo)耐受成功。 結(jié)論:①通過(guò)大鼠體內(nèi)胚胎期誘導(dǎo)免疫耐受,骨髓移植后大鼠生存狀況良好,生存時(shí)間長(zhǎng),存活率高。能夠重建造血免疫系統(tǒng),并有效的預(yù)防造血干細(xì)胞移植后的GVHD。②大鼠胚胎期體內(nèi)誘導(dǎo)免疫耐受,能夠取代大鼠移植前MHC的配型。為克服臨床造血干細(xì)胞移植須MHC配型這一限制性障礙奠定了基礎(chǔ)。③胚胎期經(jīng)卵黃囊途徑接種同種異體抗原,該實(shí)驗(yàn)方法可行。④骨髓腔內(nèi)骨髓細(xì)胞移植是一種有效的造血干細(xì)胞移植方法。
[Abstract]:Objective: to prevent GV HDD by inducing immune tolerance in rat embryonic phase, and to apply it to hematopoietic stem cell transplantation (HSCT). To overcome the restriction of MHC compatibility in hematopoietic stem cell transplantation (HSCT). Methods the recipient MHC antigen .2 receptor rats were inoculated in yolk sac at embryo stage. After irradiation with Co608.0 Gy, the general status of recipient rats was observed after transplantation of bone marrow cells in medullary cavity, and the changes of blood and bone marrow images in recipient rats were detected. The granulocytes from recipient peripheral blood were detected by PCR for GVHD related pathological examination and skin transplantation. Results the survival rate of the experimental group was 86.7 days after transplantation, which was significantly higher than that of the control group. The white blood cell count decreased significantly after 12 days and rose to normal level 20 days later. Bone marrow images showed that the bone marrow proliferated well and the donor white blood cells could be detected by PCR. The results showed that there was no GVHD phenomenon in the experimental group, and the skin grafts in the control group showed that the induced tolerance was successful. Conclusion\% 1\%\ It can reconstitute the hematopoietic immune system and effectively prevent the induction of immune tolerance in the embryonic phase of GVHD.2 rats after hematopoietic stem cell transplantation. It can replace the MHC matching before transplantation in rats. In order to overcome the restriction of MHC matching in clinical hematopoietic stem cell transplantation, the allogeneic antigen was inoculated through yolk sac at embryonic stage. This method is feasible. 4. Bone marrow cell transplantation is an effective method for hematopoietic stem cell transplantation.
