【摘要】：目的通過不同基因型的幽門螺桿菌(Hp)培養(yǎng)上清作用GES-1細(xì)胞,研究細(xì)胞Bax表達(dá)的變化。方法分組:(1)不同濃度NCTC11637Hp上清液的試驗(yàn)組,濃度分別為A組(1.3875mg/ml),B組(2.775mg/ml),C組(5.55mg/ml);(2)不同濃度NCTC11639Hp上清液的試驗(yàn)組,濃度分別為A(1.875mg/ml),B(3.75mg/ml),C(7.5mg/ml);(3)陰性對照組(Hp培養(yǎng)液);(4)空白對照組(DMEM)。各組作用GES-1細(xì)胞時(shí)間均為24h、48h、72h,用RT-PCR檢測mRNA的表達(dá)。結(jié)果 2.775mg/ml NCTC11637上清和3.75mg/ml NCTC11639上清作用GES-1細(xì)胞48h后BaxmRNA均顯著上調(diào)(P0.01~0.05)。結(jié)論不同基因型的Hp培養(yǎng)上清液可上調(diào)胃黏膜上皮細(xì)胞內(nèi)Bax的表達(dá),提示Hp可能通過激活Bax而誘導(dǎo)細(xì)胞凋亡。
[Abstract]:Objective to study the changes of Bax expression in GES-1 cells cultured with different genotypes of Helicobacter pylori (HP). Methods: (1) the supernatants of NCTC11637Hp were divided into two groups: group A (1.3875mg/ml), group B (2.775mg/ml), group C (5.55mg/ml); (2), group A (1.875mg/ml), group B (3.75mg/ml), group C (7.5mg/ml); (3), blank control group (); (4). The time of GES-1 cells in each group was 24 h, 48 h and 72 h, and the mRNA expression was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results 2.775mg/ml NCTC11637 supernatant and 3.75mg/ml NCTC11639 supernatant significantly upregulated the expression of Bax mRNA in GES-1 cells for 48 h. Conclusion the supernatant of HP culture with different genotypes can up-regulate the expression of Bax in gastric mucosal epithelial cells, suggesting that HP may induce apoptosis by activating Bax.
