【摘要】: 目的:在明確轉(zhuǎn)化生長因子β(TGF-β)誘導(dǎo)脂肪干細(xì)胞(ADSCs)向軟骨組織細(xì)胞分化作用的基礎(chǔ)上;基于轉(zhuǎn)基因技術(shù)實現(xiàn)ADSCs的TGF-β1基因修飾,研究TGF-β1在ADSCs內(nèi)表達對其分化的作用,闡明其成為理想軟骨組織工程種子細(xì)胞價值。 方法:運用TGF-β1重組腺病毒轉(zhuǎn)染ADSCs,研究ADSCs表達TGF-β1誘導(dǎo)其向軟骨組織分化作用的影響。 結(jié)果:1.轉(zhuǎn)染后ADSCs中軟骨形成相關(guān)基因的表達情況RT-PCR檢測結(jié)果顯示CollagenⅠ, CollagenⅡ, CollagenⅩ, Aggrecan在轉(zhuǎn)染后的ADSCs細(xì)胞中明顯表達增高,而ALP, PPARγ2則沒有變化。 2.GAG含量檢測:TGF-β1基因轉(zhuǎn)染的ADSCs中表達的GAG含量,隨著轉(zhuǎn)染時間的增加也顯著增加。第5,10天時,細(xì)胞內(nèi)的GAG含量與對照相比顯著增加(p0.01)。而第10天的GAG含量較第5天也顯著增加(p0.01)。 3.細(xì)胞免疫組化:TGF-β1基因轉(zhuǎn)染的ADSCs, CollagenⅡ和Aggrecan的染色均為陽性。 結(jié)論:1.TGF-β1基因轉(zhuǎn)染ADSCs內(nèi)CollagenⅠ, CollagenⅡ, CollagenⅩ, Aggrecan表達明顯增高,誘導(dǎo)其向軟骨組織分化。2.TGF-β1基因轉(zhuǎn)染ADSCs具有成為理想軟骨組織工程種子細(xì)胞價值。
