瓜蔞皮干預(yù)急性心肌梗死的藥效與機(jī)制研究

【摘要】:目的:以急性心肌梗死模型大鼠為研究對(duì)象,通過研究瓜蔞皮注射液對(duì)該模型大鼠血漿中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脫氫酶(LDH)、谷草轉(zhuǎn)氨酶(AST)、α-羥丁酸脫氫酶(α-HBDH)5項(xiàng)心肌酶的活性,內(nèi)皮素(ET)、內(nèi)皮型一氧化氮合酶(eNOS)、一氧化氮(NO)、6-酮-前列環(huán)素(6-Keto-PGF1α)、血栓素B2(TXB2)的含量,心肌中丙二醛(MDA)和超氧化物歧化酶(SOD)活性的影響,探討瓜蔞皮干預(yù)急性心肌梗死損傷的作用效果,為瓜蔞皮的臨床應(yīng)用提供實(shí)驗(yàn)依據(jù)。 方法:通過結(jié)扎大鼠左冠狀動(dòng)脈前降支,制備大鼠急性心肌梗死模型。用水合氯醛麻醉,,取血、心肌,心肌進(jìn)行切片,以氯化三苯四氮唑(TTC)染色來測定心肌梗死率;采用半自動(dòng)生化分析儀測定血漿中CK等5項(xiàng)心肌酶活性,以確定缺血心肌壞死程度;生化試劑盒檢測心肌組織中MDA及SOD活性,探討藥物清除大鼠心肌氧自由基能力;并采用ELISA法檢測NO、eNOS、ET、6-ketone-PGF1α、TXB2等指標(biāo),探討藥物對(duì)血管內(nèi)皮的保護(hù)作用和預(yù)防血栓形成的效果。 結(jié)果:與模型組相比,瓜蔞皮組可顯著降低心肌梗死大鼠心肌梗死率,維護(hù)缺血心肌正常生理功能,明顯降低CK、CK-MB、LDH、AST、α-HBDH5項(xiàng)心肌酶活性,升高心肌組織SOD活性,降低MDA含量,增加血液中NO、eNOS、6-ketone-PGF1α的含量,減少ET、TXB2的含量。 結(jié)論:瓜蔞皮對(duì)大鼠急性心肌梗死有明顯保護(hù)作用,其機(jī)制可能與抑制缺血心肌脂質(zhì)過氧化,提高組織抵抗氧自由基損傷能力,促進(jìn)血管舒張,抑制血管收縮,抑制血小板的活化、聚集,減小血栓形成等有關(guān)。
[Abstract]:Objective: to study the acute myocardial infarction model in rats. The effects of trichosanthes peel injection on the activities of creatine kinase (CK), creatine kinase isoenzyme (CK-MBN), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), 偽 -hydroxybutyrate dehydrogenase (偽 -HBDHH) in plasma of the model rats were studied. The contents of endothelin (et), endothelium-nitric oxide synthase (Enos), 6-Keto-PGF1 偽 (6-Keto-PGF1 偽), thromboxane B2TXB2 (TXB2), the activities of malondialdehyde (MDA) and superoxide dismutase (SOD) in myocardium, and the effects of trichosanthes on acute myocardial infarction injury were investigated. Methods: rat acute myocardial infarction model was established by ligating the anterior descending branch of left coronary artery. The myocardial infarction rate was determined by using chloral hydrate anesthesia, blood and myocardial sections, and the myocardial enzymes activity of CK and other 5 items in plasma were measured by semi-automatic biochemical analyzer, and the myocardial infarction rate was determined by TTC staining. In order to determine the degree of ischemic myocardial necrosis, the activity of MDA and SOD in myocardial tissue was detected by biochemical kit, and the ability of drug scavenging oxygen free radical in myocardium of rats was studied. To investigate the protective effect of drugs on vascular endothelium and the effect of preventing thrombosis. Results: compared with model group, trichosanthes skin group could significantly reduce myocardial infarction rate and maintain normal physiological function of ischemic myocardium in rats with myocardial infarction. The activity of myocardial enzymes of CK CK-MBN LDHN AST, 偽 -HBDH5 was significantly decreased, the activity of SOD was increased, the content of MDA was decreased, the content of NON-NOSS-6-ketone-PGF 1 偽 in blood was increased, and the content of ETT XB2 was decreased. Conclusion: Trichosanthes peel has obvious protective effect on acute myocardial infarction in rats. The mechanism may be related to inhibition of lipid peroxidation in ischemic myocardium, enhancement of tissue resistance to oxygen free radical damage, promotion of vasodilation, inhibition of vasoconstriction, inhibition of platelet activation, aggregation and reduction of thrombosis.
