不同濃度萬(wàn)古霉素對(duì)大鼠脂肪干細(xì)胞體外定向誘導(dǎo)分化為成骨細(xì)胞的影響

【摘要】: 目的:研究大鼠脂肪干細(xì)胞(ADSCs)在體外分離培養(yǎng)、誘導(dǎo)分化為成骨細(xì)胞及成脂細(xì)胞的能力,初步觀察不同濃度萬(wàn)古霉素對(duì)其增殖、成骨分化的影響 方法:將大鼠ADSCs放入成脂誘導(dǎo)液中培養(yǎng),采用油紅O染色觀察其成脂分化能力;同時(shí)將大鼠ADSCs放入不同濃度的萬(wàn)古霉素,以及添加了成骨誘導(dǎo)劑的培養(yǎng)液中培養(yǎng),采用MTT法、不同時(shí)相點(diǎn)測(cè)定ALP值、Von Kossa染色法觀察萬(wàn)古霉素對(duì)ADSCs的成骨影響。 結(jié)果:SD大鼠ADSCs可在不同濃度萬(wàn)古霉素溶液中生長(zhǎng),當(dāng)萬(wàn)古霉素濃度≤800μg/ml時(shí),萬(wàn)古霉素對(duì)ADSC增殖無影響,當(dāng)濃度≥1000μg/ml時(shí),才對(duì)ADSC的增殖有一定影響。萬(wàn)古霉素≤1000μg/ml時(shí),實(shí)驗(yàn)組與對(duì)照組在不同時(shí)相點(diǎn)ALP無明顯差異。通過向成脂細(xì)胞及成骨細(xì)胞定向誘導(dǎo)后,油紅染色、堿性磷酸酶染色和vonKossa染色陽(yáng)性。 結(jié)論:大鼠脂肪干細(xì)胞能向脂肪細(xì)胞和成骨細(xì)胞誘導(dǎo)分化;萬(wàn)古霉素在一定濃度范圍內(nèi)對(duì)ADSC增殖、成骨分化影響較小,當(dāng)達(dá)到某一濃度高值時(shí)才對(duì)其產(chǎn)生影響,是預(yù)防和治療骨科臨床感染的較好選擇。
[Abstract]:Objective: to study the ability of rat adipose stem cells (ADSCs) to differentiate into osteoblasts and adipoblasts in vitro, and to observe the effects of vancomycin at different concentrations on the proliferation and osteogenic differentiation of adipose stem cells. Methods: rat ADSCs was cultured in fat-forming medium, and the ability of adipogenic differentiation was observed by oil red O staining, and rat ADSCs was cultured in different concentrations of vancomycin and medium supplemented with osteogenic inducer.
