心肌缺血再灌損傷大鼠血清HSP25水平的動態(tài)變化及臨床意義初探

【摘要】: HSP25(heat shock protein 25)是小分子量熱休克蛋白之一,在細(xì)胞內(nèi)具有保護細(xì)胞骨架、抗損傷、抗凋亡等多種生物學(xué)功能。近年來研究發(fā)現(xiàn)HSP27也可以釋放到細(xì)胞外。有研究表明與健康人相比,急性冠狀動脈綜合癥病人血漿中HSP27的水平大量增加,推測其可能是一個新的心肌損傷標(biāo)志物。本研究探討心肌缺血再灌損傷時大鼠血清HSP25(與HSP27同源)的動態(tài)變化及臨床意義。 本研究首先采用結(jié)扎心臟冠狀動脈左前降支的方法制備大鼠在體心肌缺血再灌注模型,在心肌缺血30min再灌注1h、2h、4h、6h、12h、24h從心臟處取血,檢測血清HSP25、LDH、CK、CK-MB、Mb、cTnI的水平,觀察心肌缺血再灌時血清HSP25、LDH、CK、CK-MB、Mb、cTnI水平的動態(tài)變化。其次用40℃1h、40℃6h、43℃30min HS處理H9C2大鼠心肌細(xì)胞株,觀察細(xì)胞的LDH釋放率及上清液中HSP25濃度的變化。結(jié)果顯示:(1)再灌損傷組(I/R組)大鼠血清HSP25的水平從再灌注1h升高,2h達到峰值,4、6、12和24h逐漸回落,但仍高于正常水平。(2)大鼠心肌缺血再灌注損傷時,血清HSP25水平與LDH、CK、CK-MB、Mb水平具有正相關(guān)性。(3)40℃6h、43℃30minHS處理H9C2細(xì)胞株時,細(xì)胞LDH釋放率增加,上清液中HSP25的濃度升高,與對照組相比有顯著差異。上述結(jié)果提示:1.大鼠心肌缺血再灌注損傷時,小分子HSP25可以從受損心肌釋放入血,導(dǎo)致血清HSP25的水平升高。2.鑒于心肌缺血再灌注早期血清HSP25的水平上升快,敏感性高,因此血清HSP25有作為心肌損傷早期診斷指標(biāo)的潛力。
[Abstract]:HSP25 (heat shock protein 25) is one of the small molecular weight heat shock proteins. It has many biological functions such as protecting cytoskeleton, anti injury, anti apoptosis and so on. In recent years, it has been found that HSP27 can also be released to the cells. Compared with healthy people, the level of HSP27 in patients with acute coronary artery syndrome is greatly increased. In addition, it is presumed that it may be a new marker of myocardial injury. This study is to explore the dynamic changes and clinical significance of HSP25 (and HSP27 homologous) in the serum of rats with myocardial ischemia reperfusion injury.
In this study, a rat model of myocardial ischemia reperfusion was made by ligating the left anterior descending branch of the coronary artery. The blood of 1H, 2h, 4h, 6h, 12h, 24h was taken from the heart after myocardial ischemia 30min, and the serum HSP25, LDH, CK, CK-MB, Mb, and the serum levels were detected. Then, the myocardial cells of H9C2 rats were treated with 40 1H, 40 6h and 43 C 30min HS. The LDH release rate and the HSP25 concentration in the supernatant were observed. The results showed that: (1) the level of HSP25 in the serum of the reperfusion injury group (I/R group) increased from the reperfusion 1H, 2h reached the peak, and was gradually falling down, but still higher than the normal level. (2) The serum HSP25 level was positively correlated with the level of LDH, CK, CK-MB and Mb in the rat myocardial ischemia reperfusion injury. (3) at 40 C, 6h, and 30minHS treatment at 43 C H9C2 cell lines, the LDH release rate increased and the concentration of HSP25 in the supernatant increased significantly compared with the control group. The results suggested that the scores of myocardial ischemia reperfusion injury in 1. rats were small. HSP25 can be released into the blood from damaged myocardium, causing the level of serum HSP25 to rise in.2., in view of the rapid rise of serum HSP25 level and high sensitivity in the early stage of myocardial ischemia reperfusion, so serum HSP25 has the potential as an early diagnostic indicator of myocardial injury.
