【摘要】：目的:通過觀察經(jīng)前期綜合征(PMS)肝氣郁證模型大鼠下丘腦中μ-阿片受體(MOR)的蛋白表達(dá),探討舒郁膠囊治療PMS肝氣郁證的機(jī)制。方法:Wistar大鼠56只,隨機(jī)分為正常組、模型組、舒郁膠囊組(0.408 g·kg-1)和納洛酮組(0.4mg·kg-1),每組14只,曠場實(shí)驗(yàn)和陰道涂片篩選動(dòng)情周期規(guī)律非接受期大鼠進(jìn)入實(shí)驗(yàn),除正常組,其余各組采用慢性束縛應(yīng)激法復(fù)制PMS肝氣郁證模型,曠場實(shí)驗(yàn)對(duì)模型進(jìn)行行為學(xué)評(píng)價(jià),各組大鼠下丘腦采用免疫熒光化學(xué)技術(shù)(IF)和Western blot分別觀察MOR分布及蛋白表達(dá)變化。結(jié)果:宏觀行為學(xué)表明,與正常組比較,模型組大鼠曠場總分顯著降低(P0.01);免疫熒光結(jié)果顯示,與正常組比較,模型組大鼠下丘腦細(xì)胞分布排列雜亂且增多,與模型組比較,舒郁膠囊、納洛酮給藥組均得到顯著改善;蛋白免疫印跡結(jié)果顯示,與正常組比較,模型組下丘腦中MOR表達(dá)水平顯著升高(P0.01),與模型組比較,舒郁膠囊、納洛酮給藥組MOR表達(dá)水平均顯著下降(P0.01)。結(jié)論:舒郁膠囊可通過對(duì)下丘腦中MOR的拮抗作用而改善PMS肝氣郁證的癥狀,另外,下丘腦中MOR上調(diào)可能是PMS肝氣郁證的發(fā)病機(jī)制之一。
[Abstract]:Objective: to observe the protein expression of 渭 -opioid receptor moor in hypothalamus of rats with PMS-PMS-liver-qi depression syndrome, and to explore the mechanism of Shuyu capsule in treating PMS syndrome of liver-qi depression. Methods Fifty-six Wistar rats were randomly divided into normal group, model group, Shuyu capsule group (0.408 g 路kg -1) and naloxone group (14 rats in each group). Open field test and vaginal smear were used to screen non-accepted rats in estrus cycle, except the normal group. The other groups were used to establish the model of PMS liver qi stagnation by chronic restraint stress. The behavior of the model was evaluated by open field experiment. The distribution of MOR and the expression of protein were observed by immunofluorescence technique and Western blot in hypothalamus of rats in each group. Results: compared with the normal group, the total open field score of the model group was significantly lower than that of the normal group, and the immunofluorescence results showed that the distribution of hypothalamus cells in the model group was disordered and increased compared with the normal group, and compared with the model group, the results of immunofluorescence showed that the distribution of hypothalamus cells in the model group was disordered and increased compared with the model group. The results of Western blot showed that the expression of MOR in the hypothalamus of the model group was significantly higher than that in the normal group, and that in the model group, compared with the model group, the expression of MOR in the hypothalamus of the model group was significantly higher than that in the model group, and the expression of MOR in the hypothalamus of the model group was significantly higher than that in the model group. The expression of MOR in naloxone group decreased significantly (P 0.01). Conclusion: Shuyu capsule can improve the symptoms of liver qi depression syndrome of PMS by antagonistic effect of MOR in hypothalamus. In addition, upregulation of MOR in hypothalamus may be one of the pathogenesis of PMS liver qi depression syndrome.
