體外殼聚糖—明膠—生長(zhǎng)因子緩釋支架對(duì)骨髓間充質(zhì)細(xì)胞增殖的影響

【摘要】: 目的:制備具有緩釋功能的殼聚糖—明膠—堿性成纖維細(xì)胞生長(zhǎng)因子(bFGF)—肝細(xì)胞生長(zhǎng)因子(HGF)三維大孔支架,探討兩種生長(zhǎng)因子在體外對(duì)骨髓間充質(zhì)干細(xì)胞(BMSCs)增殖的協(xié)同作用。 方法:采用冷凍干燥法,將不同比例的殼聚糖、明膠、bFGF和HGF依次混合,通過冷凍、干燥,使其成為具有一定孔徑的三維緩釋支架。取SD大鼠乳鼠股骨和脛骨骨髓,分離、培養(yǎng)骨髓間充質(zhì)干細(xì)胞。取生長(zhǎng)狀態(tài)良好的第三代BMSCs,將其接種于96孔板后,加入支架混合培養(yǎng),5d后進(jìn)行MTT細(xì)胞增殖測(cè)定。實(shí)驗(yàn)過程中對(duì)動(dòng)物的處理符合倫理學(xué)標(biāo)準(zhǔn)。 結(jié)果:在與含1μg/mL的bFGF支架混合培養(yǎng),以及與同時(shí)含1μg/mL的bFGF、1μg/mL的HGF支架混合培養(yǎng)后,BMSCs明顯增殖(P0.05),但這二組間無差別(P0.05)。而與含0.1μg/mL的bFGF支架、0.01μg/mL的bFGF支架、1μg/mL的HGF支架混合后,細(xì)胞增殖無統(tǒng)計(jì)學(xué)差異(P0.05)。 結(jié)論:梯度密度離心法結(jié)合貼壁篩選法能獲得高純度高活性BMSCs;殼聚糖—明膠可作為生長(zhǎng)因子緩釋支架材料;bFGF具有促進(jìn)BMSCs增殖的作用,促進(jìn)作用的大小與加入bFGF的量有關(guān);HGF對(duì)BMSCs不具有增殖作用;bFGF和HGF在體外促進(jìn)BMSCs增殖上不具有協(xié)同性。
[Abstract]:Aim: to study the synergistic effect of two kinds of growth factors on the proliferation of bone marrow mesenchymal stem cells (BMSCs) in vitro. Methods: by freeze-drying method, chitosan, gelatin bFGF and HGF were mixed in turn to form a three-dimensional sustained-release scaffold with a certain pore diameter by freeze-drying. Bone marrow of femur and tibia were isolated from neonatal SD rats and bone marrow mesenchymal stem cells were cultured. The third generation BMSCs with good growth state were inoculated on 96-well plate and mixed with scaffold for 5 days to determine the proliferation of MTT cells. The treatment of animals in the course of the experiment accords with ethical standards. Results: after mixed culture with bFGF scaffolds containing 1 渭 g/mL and HGF scaffolds containing 1 渭 g/mL and bFGF1 渭 g/mL, the proliferation of BMSCs was significantly increased (P 0.05), but there was no difference between the two groups (P 0.05). However, when mixed with 0.1 渭 g/mL bFGF scaffold or 0.01 渭 g/mL bFGF stent or 1 渭 g/mL HGF scaffold, there was no significant difference in cell proliferation (P 0.05). Conclusion: high purity and high activity BMSCs can be obtained by gradient density centrifugation combined with adherent screening. Chitosan gelatin can be used as a growth factor sustained-release scaffold material to promote the proliferation of BMSCs. The effect of bFGF was related to the addition of bFGF to BMSCs. BFGF and HGF had no synergistic effect on the proliferation of BMSCs in vitro.
