慢性間歇性缺氧大鼠心肌氧化應(yīng)激的研究

【摘要】: 目的: 制作可應(yīng)用于常壓慢性間歇性缺氧研究的動(dòng)物模型,為進(jìn)一步研究OSAHS (Obstructive sleep apnea-hypopnea syndrome)的生理病理提供一個(gè)實(shí)驗(yàn)平臺(tái)。通過(guò)研究正常和慢性間歇性缺氧以及去除缺氧后大鼠心肌還原型谷胱甘肽(GSH)和蛋白質(zhì)羰基化(PC)的變化來(lái)探討OSAHS患者心肌是否存在氧化應(yīng)激(OS)及氧化應(yīng)激引起的進(jìn)一步損害。 方法: 選取24只SD健康成年雄性大鼠,隨機(jī)分為空白組(unhandled control, UC組)、慢性間歇性缺氧組(chronic intermittent hypoxia,CIH組)和去除缺氧組(Removal of hypoxia, RH組),UC組正常飼養(yǎng),CIH組每日間歇缺氧8小時(shí),連續(xù)4周,建立慢性間歇性缺氧模型,RH組前4周同CIH組,后4周正常飼養(yǎng),所有大鼠在實(shí)驗(yàn)結(jié)束后取心肌0.5g再提取心肌蛋白進(jìn)行免疫印跡來(lái)分析還原型谷胱甘肽(GSH)和蛋白質(zhì)羰基化(PC)的變化。 結(jié)果: ①GSH的表達(dá)CIH組較UC組和RH組明顯降低(p0.05),而UC組和RH組之間無(wú)明顯差異(p0.05)。 ②PC的結(jié)果CIH組較UC組和RH組明顯升高(p0.05),而UC組和RH組之間無(wú)明顯差異(p0.05)。 結(jié)論: 慢性間歇性缺氧致使大鼠心肌存在氧化應(yīng)激狀態(tài),并引起心肌蛋白的損傷。
[Abstract]:Purpose :



To establish an animal model applicable to the study of chronic intermittent hypoxia in normal pressure , and to provide an experimental platform for further study of the physiological and pathological features of obstructive sleep apnea - syndrome syndrome . By studying the changes of normal and chronic intermittent hypoxia and the changes of glutathione ( GSH ) and protein carbonylation ( PC ) in rats after hypoxia , it is possible to study the presence of oxidative stress ( OS ) and oxidative stress in the myocardium of patients with obstructive sleep apnea .



Method :



24 SD healthy adult male rats were randomly divided into blank group ( UC group ) , chronic intermittent hypoxia group ( group C ) , chronic intermittent hypoxia group ( CIH group ) and hypoxia group ( removal of hypoxia , RH group ) .



Results :



The expression of GSH increased significantly in CIH group than in UC group and RH group ( p < 0.05 ) , but there was no significant difference between UC group and RH group ( p < 0.05 ) .



The results of PC showed that the CIH group was significantly higher than that in UC group and RH group ( p < 0.05 ) , but there was no significant difference between UC group and RH group ( p < 0.05 ) .



Conclusion :



Chronic intermittent hypoxia causes oxidative stress in the myocardium of rats and causes damage to myocardial proteins .
