人胚胎多潛能干細(xì)胞向心肌樣細(xì)胞轉(zhuǎn)化的研究

【摘要】: 急性心肌梗死(acute myocardial infarction,AMI)是嚴(yán)重的心血管系統(tǒng)疾病,威脅著人類的健康。目前包括藥物治療、介入治療及手術(shù)治療等各種臨床治療手段,雖然在一定程度上可改善患者的癥狀,但均不能再生壞死的心肌。因此,干細(xì)胞移植成為心肌損傷治療一條很好的途徑。 本實(shí)驗(yàn)以人胚胎肝組織來(lái)源多潛能干細(xì)胞為研究對(duì)象,以5-aza為誘導(dǎo)劑,在體外將其誘導(dǎo)分化為心肌樣細(xì)胞,通過(guò)形態(tài)學(xué),RT-PCR,免疫細(xì)胞化學(xué)等方法,對(duì)誘導(dǎo)后細(xì)胞進(jìn)行鑒定,探討人胚胎肝組織來(lái)源多潛能干細(xì)胞分化為心肌樣細(xì)胞的潛能及其分化機(jī)制。結(jié)果顯示,體外人胚胎肝組織來(lái)源多潛能干細(xì)胞在適當(dāng)?shù)?-aza作用下,可分化為心肌樣細(xì)胞,RT-PCR檢測(cè)到人心肌細(xì)胞基因的表達(dá)。為今后治療心臟病藥物的研發(fā)及干細(xì)胞移植治療提供了一定的理論依據(jù)和實(shí)驗(yàn)基礎(chǔ)。
[Abstract]:Acute Myocardial Infarction (AMI) is a serious cardiovascular disease that threatens human health. At present, all kinds of clinical treatments, such as drug therapy, interventional therapy and surgical treatment, can improve the symptoms of patients to some extent, but can not regenerate necrotic myocardium. Therefore, stem cell transplantation is a good way to treat myocardial injury. In this study, multipotential stem cells derived from human embryonic liver tissue were used as the research object, 5-aza was used as inducer, the cells were induced to differentiate into cardiomyocyte-like cells in vitro, and the induced cells were identified by morphology RT-PCR, immunocytochemistry and so on. To investigate the potential and mechanism of multipotential stem cells derived from human embryonic liver tissue to differentiate into cardiomyocyte-like cells. The results showed that the multipotential stem cells derived from human embryonic liver tissue could differentiate into cardiomyocyte like cells by RT-PCR and detect the expression of human cardiomyocyte gene by RT-PCR. It provides a theoretical and experimental basis for the future development of heart disease drugs and stem cell transplantation therapy.
