【摘要】： 目的:選取Wistar大鼠為實(shí)驗(yàn)對(duì)象,通過(guò)建立慢性牙周炎和動(dòng)脈粥樣硬化結(jié)合的動(dòng)物模型并進(jìn)行相關(guān)性的初步分析,探討慢性牙周炎和動(dòng)脈粥樣硬化發(fā)病相關(guān)的可能機(jī)制。方法:雄性Wistar大鼠(體重200±20g)60只隨機(jī)分為四組:A對(duì)照組,B慢性牙周炎組,C動(dòng)脈粥樣硬化組,D慢性牙周炎+動(dòng)脈粥樣硬化組。各組按照實(shí)驗(yàn)設(shè)計(jì)進(jìn)行相應(yīng)干預(yù)處理,至12周末處死動(dòng)物,分別檢查記錄牙周指標(biāo),檢測(cè)血清中的血脂水平,酶聯(lián)免疫吸附法(ELISA)檢測(cè)血清中的腫瘤壞死因子α(TNF-α)以及基質(zhì)金屬蛋白酶-3(MMP-3)的含量,免疫組化技術(shù)分別檢測(cè)MMP-3在牙周和腹主動(dòng)脈組織中的表達(dá),并進(jìn)行圖像半定量分析,同時(shí)觀察牙周和腹主動(dòng)脈的組織病理學(xué)改變。數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果:經(jīng)過(guò)牙周檢查、血脂、ELISA檢測(cè)以及組織病理學(xué)觀察,已成功建立了動(dòng)物模型。其中D組的血脂水平、血清TNF-α及MMP-3含量明顯升高,各實(shí)驗(yàn)組與對(duì)照組比較差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。經(jīng)過(guò)免疫組化檢測(cè)發(fā)現(xiàn),MMP-3在B組和D組的牙周組織中,主要在牙齦上皮呈陽(yáng)性表達(dá),并且D組水平高于B組;在實(shí)驗(yàn)組的腹主動(dòng)脈中,主要在內(nèi)皮細(xì)胞、巨噬細(xì)胞、平滑肌細(xì)胞呈陽(yáng)性表達(dá),并且D組水平高于C組,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。MMP-3在組織中的表達(dá)與血清中的規(guī)律相一致。MMP-3與血脂的相關(guān)性分析呈現(xiàn)出不相關(guān)。結(jié)論:慢性牙周炎與動(dòng)脈粥樣硬化發(fā)病可能具有一定的相關(guān)性,可能對(duì)動(dòng)脈粥樣硬化和冠心病的發(fā)生發(fā)展有重要的影響。MMP-3是獨(dú)立于血脂之外的動(dòng)脈粥樣硬化危險(xiǎn)因素。
[Abstract]:Objective: to investigate the possible mechanism of chronic periodontitis and atherosclerosis by establishing the animal model of chronic periodontitis and atherosclerosis in Wistar rats. Methods: male Wistar rats (body weight 200 鹵20g)60) were randomly divided into four groups: control group (n = 4) and chronic periodontitis group (n = 4). According to the experimental design, the animals were killed at the end of 12 weeks. The periodontal indexes were recorded and the serum lipids were measured. Serum levels of TNF- 偽 and matrix metalloproteinase-3 (MMP-3) were detected by Elisa, and the expression of MMP-3 in periodontal and abdominal aorta was detected by immunohistochemistry. The histopathological changes of periodontal and abdominal aorta were observed. The data were analyzed statistically. Results: the animal model was successfully established by periodontal examination, Elisa and histopathological observation. The levels of blood lipid, TNF- 偽 and MMP-3 in group D were significantly higher than those in control group (P 0.05). The expression of MMP-3 in the periodontal tissues of group B and group D was mainly positive in gingival epithelium, and the level of MMP-3 in group D was higher than that in group B. in the abdominal aorta of experimental group, the expression of MMP-3 was mainly in endothelial cells and macrophages. The expression of smooth muscle cells was positive and the level of group D was higher than that of group C. the difference was statistically significant. The expression of P0.05U. MMP-3 was consistent with the rule of serum. There was no correlation between MMP-3 and serum lipids. Conclusion: chronic periodontitis may be associated with the pathogenesis of atherosclerosis, which may have an important effect on the occurrence and development of atherosclerosis and coronary heart disease. MMP-3 is a risk factor of atherosclerosis independent of blood lipids.
